摘 要:目的:研究注射用重组人甲状旁腺素(1-34)在健康人体内的药动学特征,并进行了药效学考察。方法:10名健康受试者,男女各半,分别单次和多次皮下注射重组人甲状旁腺素(1-34)40μg。采用放射免疫分析法检测血浆中重组人甲状旁腺素(1-34)的浓度,用DAS2.1.1软件进行数据处理,计算药动学参数,并用SPSS软件对其中的结果进行统计分析。结果:10名健康受试者分别单次、多次皮下注射重组人甲状旁腺素(1-34)40μg的主要药动学参数如下:tmax为(45.0±10.0)min和(51.0±16.1)min;Cmax为(231.3±87.4)pg·mL-1和(247.2±92.3)pg·mL-1;AUC(0-240)为(27 379.5±9 080.2)pg·min·mL-1和(29 373.3±5 760.6)pg·min·mL-1;AUC(0-∞)为(33 142.0±9 890.9)pg·min·mL-1和(37 311.1±9 015.6)pg·min·mL-1;t1/2为(86.1±51.8)min和(94.8±53.6)min;Vz为(155.7±73.2)L和(144.7±60.7)L;CLz为(1.3±0.4)L·min-1和(1.1±0.3)L·min-1。结论:注射用重组人甲状旁腺素(1-34)单次与多次皮下注射给药的药动学参数差异无显著性意义(P>0.05),连续给药后在体内无蓄积。性别对注射用重组人甲状旁腺素(1-34)的体内药动学影响无差异。OBJECTIVE To study the pharmacokinetics and pharmacodynamics of recombinant human parathyroid hormon(1-34) for injection in Chinese healthy subjecls. METHODS A total of 10 healthy volumeers (5 males and 5 females) were ad ministrated with single and muhiple dose of recombinant human parathyroid hormone (1-34) 40μg, respectively. The concen trations of rhPTH(1 34) in plasma were determined by IRMA while the phannacokinetic parameters were calculated by DAS vet 2. 1. 1 software. The statistical package SPSS was used for all statistical analysis. RESULTS The main pharmacokinelic parameters of reeombinant human parathyroid hormone ( 1 34) after single and multiple-dose subcutaneous administration were as follows: t = (45.0-± 10.0) and (51.0±16. 1)min;(231.3±87.4) and (247.2±92.3)pg.mL^-1; AUC(0-240)= (27379.5±9080.2) and (29373.3±5 760.6)pg.min.mL^-1; AUC(0-∞)= (33 142.±9 890.9) and (37 311.1 ±91)15.6) pg·min·mL^-1; t1/2 =(86.1±51.8) and (94.8±53.6)min; Vz= (155. 7 ± 73. 2) and (144.7±60.7) L; CLz =(1.3±0.4) and (1. 1 ± 0. 3)L·min-1, respectively. CONCLUSION After multiple doses, the pharmacokinetic parameters of recombinant hu man parathyroid hormone (1-34) are consistent with those derived after a single dose, indicating no significant accumulation of recombinant human parathyroid hormone (1-34) with repeated dosing. Gender appears to have no significant effect on the phar macokinetic properties of recombinant human parathyroid hormone (1-34).
