四藤方对人胃癌裸鼠皮下移植瘤生长及凋亡作用

摘　　要：目的:观察清热解毒中药四藤方对人胃癌SGC-7901细胞裸鼠移植瘤生长及凋亡影响。方法:人胃癌SGC-7901细胞BALB/C裸小鼠皮下移植瘤模型,随机分为对照组、四藤方组、5-Fu组3组,每组10只;对照组予0.9%氯化钠溶液0.3mL灌胃,1次/d;四藤方组予四藤方煎剂0.3mL灌胃,1次/d;5-Fu组予5-Fu 1mg腹腔注射,每周1次,每次1mg;观察各组裸小鼠移植瘤生长情况。TUNEL法检测细胞凋亡,免疫组织化学法检测肿瘤组织凋亡相关蛋白半胱天冬酶-8(Caspase-8)、Caspase-9表达情况。结果:四藤方组、5-Fu组肿瘤生长变缓,裸小鼠移植瘤体积、质量与对照组比较差异有统计学意义(P<0.05,P<0.01);TUNEL检测显示四藤方组、5-Fu组移植瘤细胞凋亡率高,与对照组比较差异有统计学意义(P<0.01);四藤方组与5-Fu组治疗后移植瘤组织Caspase-8、9蛋白表达升高,与对照组比较差异有统计学意义(P<0.05,P<0.01)。结论:四藤方可以抑制SGC-7901胃癌肿瘤生长,可能与促进细胞凋亡,上调凋亡相关蛋白Caspsse-8、9表达相关。Objective： To investigate the effects of Siteng Fang, a Chinese herbal formula, on tumor growth and apoptosis in gastric carcinoma. Methods： 30 BALB/C nude mice subcutaneously transplanted with SGC-7901 cells were randomly divided into three groups （10 mice per group）, and intragastric administered with Siteng Fang or normal saline daily, or i.p. administered with 5-Fu weekly. Tumor volume was observed. Cell apoptosis was detected by TUNEL assay. Expression of Caspase-8 and 9 in the tumor tissue were detected by immunohistochemistry. Results： Siteng Fang and 5-Fu significantly reduced tumor volume, quality and compared with controls （P〈0.05, P〈0.01）, accompanied by significant apoptosis in human gastric carcinoma xenografts （P〈0.01）. The expression of Caspase-8 and 9 in tumor tissue were significantly up-regulated by Siteng Fang 5-Fu treatment （P〈0.05, P〈0.01）. Conclusion： Siteng Fang may inhibit tumor growth and promote cell apoptosis in SGC-7901 gastric carcinoma, and may relate to Caspsse-8 and 9 expression.
