摘  要:目的:通过观察失眠大鼠下丘脑内谷氨酸(Glu)、γ-氨基丁酸(GABA)的含量及γ-氨基丁酸A型受体(GABAA)阳性表达的变化,探讨电针治疗失眠的可能机制。方法:采用对氯苯丙氨酸(PCPA)制备失眠模型,观察各组大鼠的行为反应,Glu、GABA的含量及GABAA受体的阳性表达,进而探讨电针治疗失眠的部分机制。结果:治疗后,与模型组比较,电针组大鼠Glu、Glu/GABA比值明显降低(P<0.05),GABA含量明显升高(P<0.05),GABAA受体阳性细胞数增加;药物组大鼠脑内Glu、GABA含量及其比值无明显差异,GABAA受体阳性细胞数增多。结论:针刺与安定治疗失眠均有疗效,且针刺无副作用,优于安定,具有调节脑内Glu、GABA含量以及GABAA受体活性的多靶性综合作用。Objective: By observing the influence of electro-acupuncture on the content of glutamic acid and aminobutyric acid, and the positive expression of GABAA receptor in the brain of rats with insomnia, to explore the possible mechanism of electro-acupuncture on insomnia. Methods: The insomnia model was induced by PCPA peritoneal injection. To observe therapeutical effect of electro-acupuncture on rat model according to the rotational behavior, the content of Glu and GABA in the rat's hypothalamus, and the positive expression of GABAA receptor. Results: Compared with the model group, after treatment of electro-acupuncture, the content of GIn and the rate of Glu/GABA decreased obviously (P〈0.05), the content of GABA and the immunohistochemical positive cells of GABAA receptor increased (P〈0.05). Compared with the model group, after the treatment of diazepam,contents of Glu and GABA and their rate were not significantly different, and positive cells of GABAA receptor were increased. Conclusions: The sleep-waking cycle of rats with insomnia caused by PCPA can be renewed by both electro-acupuncture and Diazapan treatment, but electro-acupuncture is superior to Diazepan in the treatment of insomnia due to its good regulation on neurotransmitters and their receptors.
