摘 要:目的:观察灯盏花素对大鼠液压冲击伤性脑水肿水通道蛋白4(AQP4)表达的影响。方法:成年SD大鼠80只,应用Dixon法制作脑损伤模型,随机分为两组:模型组和干预组,各40只。干预组应用灯盏花素90mg/kg腹腔注射。两组动物均于术后6、12、24h、3、7d进行神经功能评分和组织学观察,并用干湿重法、免疫织化法分别测定水肿脑组织含水量及其AQP4表达。结果:与模型组比较,干预组大鼠脑组织水肿程度减轻,脑组织含水量则以24h、3d减轻最为明显(P<0.05);神经功能评分出现明显改善,以12h、24h、3d、7d 4个时间点最为显著(P<0.05),同时AQP4表达也出现不同程度减少,尤以12h、24h、3d、7d为著(P<0.05)。结论:灯盏花素能够通过下调AQP4表达减轻液压冲击脑损伤脑水肿,发挥神经保护作用。Objective: To investigate the effect of Breviscapine on AQP4 expression of fluid percussion injury in rats. Methods: 80 rats were employed to establish experimental models according to Dixon's method and randomly divided into two groups: model group and intervention group, the later of which were adminstered Breviscapine (90mg/kg) by way of intraperitoneal injection. Nervous function score, brain water content, histological changes, AQP4 expression were observed by Shapira and Wahld method, dry-wet measure, light microscopy, immunohistochemistry at 6, 12, 24h, 3d and 7d after strike respectively. Results: Compared with animals of model group, the pathological changes and water content of edemamtous brain in those of intervention group significantly improved, especially in 24h and 3d after strike (P〈0.05). Nervous function score increased significantly, especially in 24h, 3d and 7d after strike (P〈0.05). AQP4 immunoreactive expression decreased signifacantly in 24h, 3d and 7d after strike (P〈O.05). Conclusion: Breviscapine plays a role of neuroprotective effect on brain damage from fluid percussion injury by downregulating AQP4 expression.
