摘 要:目的:探讨加味丹栀逍遥散对抑郁模型大鼠下丘脑内5-HT和NE含量的影响。方法:将50只Wistar雄性大鼠随机分为对照组,模型组,氟西汀组,加味丹栀逍遥散高剂量组、低剂量组,每组各10只;采用慢性轻度不可预见性应激结合孤养的方式复制复合模型,除对照组外,其余各组均接受不同应激因子刺激21 d,建立慢性应激抑郁大鼠模型。氟西汀组,加味丹栀消遥散高剂量组、低剂量组分别按2.5 mg·kg-1、4.08 g·kg-1、2.04 g·kg-1剂量给药,各组均按10 mL·kg-1灌胃给药,对照组和模型组按10 mL·kg-1给予生理盐水每日1次,应激前灌胃,从造模第22天开始,至动物处死当天,共21 d,实验周期共42 d;采用酶联免疫法(ELISA)测定大鼠下丘脑内5-羟色胺(5-HT)和去甲肾上腺素(NE)含量的变化。结果:与对照组比较,模型组大鼠下丘脑内5-HT和NE含量明显减少;经过治疗后,氟西汀组与加味丹栀逍遥散高剂量组均显著高于模型组(P<0.05),加味丹栀逍遥散低剂量组NE含量显著高于模型组(P<0.05)。结论:加味丹栀逍遥散抗抑郁作用可能与升高下丘脑内5-HT和NE的含量有关。Objective :To investigate the effect of modified Danzhi Xiaoyao powder on 5-HT and Norepinephrine(NE) in hypothalamus of depression model in rats. Methods : Fifty male Wistar rats were randomly divided into blank control group, model group, fluoxertine hydroehloride group, modified Danzhi Xiaoyao powder low dose and high dose group, and 10 in each group. Depression model of rat was produced by using chronic unpredictable mild stress stimulation combined with lonely raising for 21 days. From the twenty-second days of models, fluoxertine hydrochloride group 2.5 mg ~ kg - 1, modified Danzhi Xiaoyao powder low dose group 2.04 g ~ kg - l, high dose group 4.08 g ~ kg-l ,each of the medication group perfused daily with the medicine which was needed per 10 mL ~ kg-1 ,and blank control group and model group perfused daily with normal saline of a volume the same as the medication group, before the stress of each day. Until all rats were killed,there were 21 days. The experimental period was a total of 42 days. Determined the change of 5-hydroxy tryptamine (5-HT) and NE in hypothalamus of rats by euzymelinked immunosorbent assay(ELISA). Results:The eoncentraions of 5-HT and NE in hypothalamus of the model rats were both reduced significantly compared to that of the blank control group( P 〈 O. 05 ) ;After treatment, the concentraions of 5-HT and NE in hypothalamus of modified Danzhi Xiaoyao powder high dose group were increased signif- icantly compared to that of the model group (P 〈 0~ 05 ), and just the concentraion of NE of modified Danzhi Xiaoyao powder low dose group was significantly higher than that of the model group. Conclusion : The mechanism for anti-depression of modified Danzhi Xiaoyao powder may be correlative to regulating the coneentraion of NE and 5-HT in hypothalamus of depression model in rats.
