摘  要:目的:探讨地黄饮子对急性脑缺血大鼠神经干细胞迁移的影响。方法:采用大脑中动脉线栓法建立右侧局灶性脑缺血大鼠模型,将大鼠随机分为模型组、假手术组、中药组、尼莫地平组。假手术组只打开颈部皮肤,毕露颈部血管后不做其他任何处理即缝合;地黄饮子组动物于术后第4天起按4 mL·kg-1灌服地黄饮子药液,每日1次,给药14 d;尼莫地平组每天按1mg·kg-1给予尼莫地平;模型组及假手术组均给予等体积无菌蒸馏水,自由饮食、活动。在第4天至第7天,第18天至第21天肌肉注射5-溴脱氧尿嘧啶核苷(BrdU)50 mg·kg-1,1次·d-1,之后处死动物取材。治疗第1周、第2周、第3周后进行神经功能评分,并检测细胞基质衍生因子(SDF-1)蛋白含量。结果:在第1周末,与模型组比较,尼莫地平组、地黄饮子组神经功能评分和SDF-1含量均降低,但差异无统计学意义(P>0.05)。在第2周、第3周末地黄饮子组显著低于模型组和尼莫地平组,差异有统计学意义(P<0.05)。结论:地黄饮子可通过降低脑缺血大鼠脑内SDF-1含量,进一步促进内源性神经干细胞迁移。Objective : To explore the influence of Dihuang Yinzi on migration of neural stem cells of cerebral ischemia rats. Methods : U- sing middle cerebral artery occlusion to establish the right focal cerebral ischemia model in rats, and the rats were randomly divided into the model group, sham-operation group, Chinese medicine group and nimodipine group. In the sham-operation group, only opened the neck skin of rats, exposed cervical blood vessels and didn't do any processing and immediately stitched. Dihuang Yinzi group was given drenching Dihuang Yinzi liquid according to 4 mL ~ kg-1 at the 4th day postoperative, 1 time a day, and medication for 14 days. The model group and sham-operation group both were given the same volume of sterile distilled water,freely diet and activities, on the 4th day to the 7th day, and the 18th day to the 21st day, the rats were given intramuscular 5-bromodeoxyuridine (BrdU) ,50 mg ~ kg-1,1 time a day, and the animals were sacrificed for materials. The neurological function was scored in the treatment of 1,2,3 weeks, and the protein content of cell matrix derived factor (SDF-1) was detected. Result:In the first weekend, nimodipine group and Dihuang Yinzi both can reduce the neurological function score and the content of SDF-1 and there was no significant difference compared with the mod- el group (P 〉 O. 05 ). In the 2nd and 3rd weekend, the neurological function score and the content of SDF-1 of Dihuang Yinzi group was significantly lower than that of model group and nimodipine group, and the difference was statistically significant ( P 〈 0.05 ). Conelu- sion:Dihuang Yinzi can reduce SDF-1 content of cerebral ischemia rats, and further promote the migration of endogenous neural stem cells.
