心得宁口服液的急性毒性实验

摘　　要：目的:观察心得宁口服液对小鼠急性毒性的作用。方法:40只小鼠按随机数字表平均分为心得宁口服液组和对照组。实验前禁食、不禁水12 h,心得宁口服液组灌服最大可灌服浓度的心得宁口服液混悬液,每天3次,每次间隔8 h;对照组灌服同体积的生理盐水。两组小鼠均正常饮食,连续观察14 d。其每日积累最大耐受量为72 g·kg-1,该量已为临床用量的1 200倍,小鼠未出现死亡,处死小鼠解剖。结果:心得宁口服液组小鼠解剖肉眼未见肝、心、脾、肺、肾、胃、肠、脑等有明显异常。心得宁口服液组小鼠第1天体质量为(20.0±0.9)g,第7天体质量为(25.2±1.6)g,第14天体质量为(27.7±1.7)g;生理盐水对照组小鼠第1天体质量为(19.8±0.8)g,第7天体质量为(25.1±1.4)g,第14天体质量为(27.5±1.7)g。结论:心得宁口服液混悬液口服毒性很小,临床用药安全。Objective：To observe the acute toxicity effect of Xindening Oral Liquid on mice. Methods：40 mice with body weight of 19 - 22 g, female and male half and half, were randomly assigned to the xindening oral liquid group and the control group. Not-deprived of water, and fasting 12 hours before the experiment,xindening oral liquid group was drenched the maximum volume drenching concentra- tion of Xindening Oral Liquid suspension,3 times a day, each time interval 8h;the control group was drenched with the same volume of saline solution. And the two groups of mice were given normal diet, and continuous observation for 14 days. The daily accumulated Maxi- mum Tolerated Dose was 72 g · kg-1, the dose was 1200 times of the clinical dosage, and death didn＇t occur in mice, mouse anatomy death. Results ： mice can drench the maximum concentration in the greatest volume drench intragastric, not visible to the naked eye of lung, kidney,liver, heart, spleen, stomach, intestine, brain, and other obvious exceptions. Body weight of mice of xindening oral liquid group was （20.0 ± 0.9） g in the beginning,and（25.2 ± 1.6） g on the 7th day,and （27.7 ± 1.7 g） on the 14 th day;body weight of mice of physiological saline control group was （ 19.8 ± 0.8） g in the beginning, and（25.1 ± 1.4） g on the 7th day, and（27.5 ± 1.7） g on the 14th day. Conclusion：Oral administration Xindening Oral Liquid suspension has small toxicity, and safe clinical medication.
