摘  要:目的:从针灸对环磷酰胺(CTX)化疗小鼠骨髓造血微环境的影响,探讨针灸改善化疗后骨髓抑制,提升外周血白细胞的作用机制。方法:选用昆明种清洁级小鼠64只,称质量后依据体质量随机分为空白组、模型组、针刺组、艾灸组4组,每组8只。空白组小鼠腹腔注射生理盐水,剩余3组小鼠腹腔注射CTX溶液造成骨髓抑制模型。造模完成4 h后,针刺组、艾灸组小鼠分别选取大椎、膈俞、肾俞、足三里施以针刺和艾灸,空白组、模型组陪同固定。同时分别在造模前、造模后及治疗3 d、5 d后割尾取静脉血,检测外周血白细胞,并分批处死小鼠取股骨内骨髓组织,用免疫组织化学法检测骨髓组织中细胞间黏附分子(ICAM-1)、血管细胞黏附分子(VCAM-1)蛋白表达。结果:针刺、艾灸均可以提高CTX化疗小鼠骨髓组织中黏附分子ICAM-1、VCAM-1蛋白表达,升高外周血白细胞。结论:针灸提高CTX化疗小鼠骨髓组织中黏附分子ICAM-1、VCAM-1蛋白表达,利于造血细胞与组织细胞之间的黏附和信息交换,增强造血细胞对细胞因子反应的敏感性,促进造血细胞的分裂、增殖、释放,应当是针灸参与机体造血调控,修复骨髓造血微环境的损伤,改善化疗后骨髓抑制,提升白细胞的重要机制之一。Objective:To explore the mechanism of acupuncture and moxibustion on improving marrow supression after chemical therapy and enhancing leukocytes in peripheral blood by observing the influence of acupuncture and moxibustion on marrow hematopoietic mi- croenvironment in post CTX chemotherapy mice. Methods : Selected 64 clean level kunming mice and then randomly divided into normal group, model group,acupuncture group and moxibustion group according to body weight, each of 8 mice. The normal group was injected with NS, CTX was used for the rest three groups, then the models of myelosuppression were estabished 4h post repeated injections. Se- lected points of "Dazhui" ," Geshu " ," Shenshu" and "Zusanli" ,then applied acupuncture and moxa to the mice 4h after modeling in the corresponding groups, normal group and model group accompanied fixed, at the same time, before modeling, after modeling and 3 days,5 days after treatment,cutting the tails to obtain blood and detected the leukocytes counting in peripheral blood respectively, then killed a batch of mice and taking out bone marrow tissue of thighbone,and observed the expression of adhesion molecules as ICAM-1, VCAM-1 with immunohistochemical method. Results:Acupuncture and moxibustion can both promote marrow expression of VCAM-1 and VCAM-1 in CTX-induced mice, and elevate the leukocytes in peripheral blood. Conclusion:Acupuncture and moxibustion can both promote marrow expression of VCAM-1 and VCAM-1 in CTX-induced mice, benefit the adhesion and information exchange between hematopoietic cells and tissues, enhance sensibility of hematopoietic cells to cytokine, promote cell division, proliferation and release of hematopoietic cells,which shoud be one. of the mechnisms of acupuncture and moxibustion regulating hematopoietic function, repairing marrow injury of hematopoietic py with CTX. , and then elevate the leukocytes, relieving myelosuppression in mice post chemothera-
