芪术胶囊对糖尿病肾病大鼠肾组织TGF-β1和CTGF表达的影响

摘　　要：目的观察芪术胶囊对糖尿病肾病(DN)大鼠肾脏保护作用。方法采用单侧肾脏切除和腹腔注射链脲佐菌素的方法来建立大鼠糖尿病肾病模型,随机分为正常(NC)组、模型组(DM)、芪术胶囊(QZ)组、氯沙坦钾(LSTJ)组;成模1周后,QZ组按1 g·kg-1给药,LSTJ组按1.67 mg·kg-1给药。实验8周末观察各组大鼠血尿素氮BUN、血肌酐(Scr)、糖化血红蛋白(HbAlc)、胱抑素(CysC)以及24 h尿蛋白定量,并进行肾组织病理形态学观察以及免疫组化法检测肾组织中转化生长因子β1(TGF-β1)与结缔组织生长因子(CTGF)的蛋白表达。结果与DM组相比:QZ组大鼠血清BUN、Scr、HbAlc、Cys-c和24 h尿蛋白定量明显减低,肾组织病理形态学改变程度均明显改善,大鼠肾小球TGF-β1和CTGF表达均减弱。结论芪术胶囊可以减少尿蛋白,降低血糖,改善肾功能,下调肾组织TGF-β1和CTGF表达,起到保护肾脏,延缓DN进展的作用。Objective To observe the kidney protective function of Qizhu Capsules in the rats of di- abetic nephropathy （DN）. Methods The unilateral nephrectomy and the intraperitoneal injection of Strepto- zotocin （STZ）were adopted to build DN rat model. A normal group, a model group, a Qizhu （QZ）group and a Losartan Potassium（LP） group were randomized. In 1 week of modeling, Qizhu Capsules were given at 1 g · kg- i in the QZ group and Losartan Potassium was prescribed at 1.67 mg· kg - 1 in the LP group. In 8 weeks of experiment,the levels of BUN, Scr, HbAlc, CysC and 24 h urinary protein were observed in the rats of each group. The kidney tissue was observed in pathomorphology. IHC assay was used to determine the expressions of TGF - β1 and CTGF in kidney tissue. Results Compared with the model group,the levels of BUN, Scr, HbAlc, CysC and 24 h urinary protein in serum were reduced obviously in the QZ group. The pathomorpholog- ical changes were relieved apparently in kidney tissue. The expressions of TGF - β1 and CTGF of glomeruli were all decreased in the rats. Conclusion Qizhu Capsules decrease the levels of urinary protein and blood glucose,improve kidney functions and down -regulate the expressions of TGF- β1 and CTGF in kidney tis- sue. This medication protects the kidney and delays the progression of DN.
