摘 要:目的 从伤口早期中性粒细胞 (Neu)肌动蛋白 (actin)的变化方面探讨全身γ射线照射后Neu吞噬、趋化功能变化的机理及促愈药康复新 (W11 a12 )的作用机理。方法 以大鼠 6Gy全身照射复合皮肤切割伤为模型 ,收集伤口细胞。用Fluorescein 鬼笔环肽标记NeuF actin进行测定和观察。结果 复合伤组NeuF actin含量先高后低 ,对FMLP反应性减弱 ,FMLP刺激后解聚慢 ,康复新能改善NeuF actin的功能状态。结论 电离辐射引起Neu肌动蛋白功能异常可能是Neu趋化、吞噬活性降低的重要原因 ,改善NeuF actin的功能状态是康复新提高Neu吞噬和趋化功能的作用机理。Objective\ To explore the mechanism of decreased phagocyotic and chemotactic function of neutrophils (Neu) and the mechanism of W 11 \|a 12' s pharmacological action by studying the changes of actin of Neu in wound after total body irradiation of rats. Methods\ Cells were collected from incision wounds on back of rats after 6 Gy whole body\|irradiation.F\|actin in Neu was stained with Fluorescein\|phalloidin. Results\ There were increased (3~12 h) and decreased (24 h,48 h) basal levels of F\|actin,a lower responsiveness to stimulation and a reduced rate of actin depolymerization after combined radiation injury compared with simple wound group,which were important causes of Neu dysfunction. Conclusion\ W 11 \|a 12 could improve the functional state of Neu actin,which may contribute to the improved chemotactic and phagocytotic function of Neu after administration of this drug.
