12-脂加氧酶抑制剂黄芩素对病毒性心肌炎小鼠动物模型的干预作用研究

摘　　要：目的 探讨12-脂加氧酶（12-LO）抑制剂黄芩素对病毒性心肌炎小鼠动物模型的干预作用.方法 将250只4周龄BALB/C雄性小鼠采用计算机随机分组的程序随机分为5组,正常小鼠空白对照组（A 组）、模型组（B组）、高剂量治疗组（C组,0.4 g/L黄芩素溶液）、中剂量治疗组（D组,0.2 g/L黄芩素溶液）和低剂量治疗组（E组,0.1 g/L黄芩素溶液）,每组50只;分别于病毒接种后第4、7、14、21天随机取部分小鼠观测心肌组织病理学改变（HE染色法）、心肌组织中12-LO mRNA表达（RT-PCR法）及血清肌酸激酶同工酶（CK-MB）水平（化学发光免疫法）.采用SPSS 13.0统计软件对5组小鼠的心肌组织病理损害情况、12-LO mRNA表达量及血清CK-MB水平进行统计学分析.结果 5组小鼠各时间段的心肌组织病理积分比较,差异均有统计学意义（P＜0.05）,其中B组小鼠于病毒接种后第4、7、14、21天心肌组织病理积分均高于A组 （P＜0.01）,C、D、E组小鼠各时间段心肌组织病理积分均低于B组（P＜0.05）.B组小鼠柯萨奇B3病毒（CVB3）接种后第4天CK-MB水平即有改变,第7～14天达到高峰,第21天降低,与A组相比,其中第4、7、14天差异有统计学意义（P＜0.05）.C、D、E组小鼠第4、7、14天的CK-MB水平均低于B组相应时间段的水平（P＜0.05）,但仍未降到正常水平.不同时间段各组小鼠心肌组织12-LO mRNA相对表达量比较,差异均有统计学意义（P＜0.05）,其中B组小鼠于CVB3接种后第4、7、14、21天心肌组织12-LO mRNA相对表达量均高于A 组（P＜0.01）;C、D、E组小鼠心肌组织12-LO mRNA相对表达量除第4天外,其他各时间点的表达量均低于B组（P＜0.05）.各治疗组12-LO mRNA的表达量与心肌组织病理积分及血清CK-MB水平均呈正相关（P＜0.05）.结论 12-LO抑制剂黄芩素可能通过降低CVB3感染后小鼠心肌组织12-LO的表达,减轻病毒性心肌炎病情,为开发治疗病�Objective To investigate the effect of 12 - LO inhibitor baicalein in the intervention on mice model of viral myocarditis. Methods 250 four - week - old BALB/C male mice were randomly divided into 5 groups, namely group A （ blank control group with normal mice）, group B （model group）, group C （high dose treatment group, O. 4 g/L baicalein solution）, group D （middle dose treatment group, 0. 2 g/L baicalein solution） and group E （low dose treatment group, 0. 1 g/L baicalein solution）, with each group 50 mice. On the 4th, 7th, 14th and 21st day after virus inoculation, myocardial tissues of some mice were randomly obtained to observe pathological change, 12 -LO mRNA and CK- MB in myocardial tissue by HE staining, RT - PCR and chemiluminescence immunoassay methods respectively. The tissue pathological damage, 12 - LO mRNA expression and serum levels of CK- MB were statistically analyzed by SPSS 13.0 statistical software. Results The differences of myocardial histopathology score in each group were statistically significant （ P 〈 0.05 ） . Each time the myocardial histopathology score of group B were higher than the one of group A, and the difference was statistically significant （ P 〈 O. 01 ） ; the myocardial histopa- thology score of C, D, E groups in each time were lower than group B, and the differences was statistically significant （ P 〈 0.05 ） . The CK - MB level of group B mice changed on the 4th day after CVB3 inoculation, and reached the peak level on the 7th to 14th day, and reduced on the 21st day. Compared with group A, the difference of CK - MB level on the 4th, 7th and 14th days was statistically significant （P 〈 0. 05） ; The CK - MB levels of C, D, E groups at each time were lower than the cor- responding period in group B levels （P 〈 O. 05 ）, but did not drop to normal levels.
