摘 要:目的建立液相色谱串联质谱(LC-MS)测定方法并检测姜黄素的大鼠血浆蛋白结合率。方法建立LC-MS检测方法,结合超滤法测定姜黄素的血浆蛋白结合率。结果3种浓度(100、300和1000ng·mL^-1)姜黄素与大鼠血浆蛋白结合率分别为(80.3±5.28)%、(75.3±4.45)%和(62.64—1.98)%。结论姜黄素血浆蛋白结合率较高,LC—MS可用于姜黄素血浆蛋白结合率的测定。OBJECTIVE To establish the method with liquid chromatography-mass spectrophotometer(LC-MS) and determine the protein binding rates of curcumin in plasma of rat. METHODS The ultrafiltration combined with LC-MS was performed to determine the drug concentration of curcumin, and its protein binding rates in rat. RESULTS With different concentration of curcumin including 100,300 and 1000ng ~ mL-1 , the protein bind- ing rates of curcumin in rat were ( 80. 3 ± 5.28 ) %, ( 75.31± 4, 45 ) % and (62. 6 ± 1.98 ) %. CONCLUSION curcumin has higher level in binding to plasma protein of rat. The LC-MS method can be used to determine the drug concentration of curcumin properly.
