Determination of Palmatinechloride and Berberine Hydrochloride In Coptis chinensis Franch by HPLC and Fluorescence Detection

摘  要:目的采用高效液相荧光法建立测定黄连中盐酸巴马汀和盐酸小檗碱的方法。方法色谱柱AtlantisC18(2.1mm×100mm,3μm),乙腈.15mmol·L^-1十二烷基硫酸钠磷酸盐缓冲液(0.05mol·L^-1磷酸二氢钾,用磷酸调节pH=4.0)(40:60),荧光检测器激发波长为354nm,发射波长为533nm。结果盐酸巴马汀浓度在1.798—179.8μg·mL^-1。范围内与峰面积呈良好的线性关系,r为1.000;盐酸小檗碱浓度在I.762—176.2μg·mL-1范围内与峰面积呈良好的线性关系,r为0.9995;盐酸巴马汀加样回收率的平均值为99.98%,RSD=3.8%;盐酸小檗碱加样回收率的平均值为99.12%,RSD=1.4%。结论本方法简便、准确可靠,适用于黄连中盐酸巴马汀和盐酸小檗碱的质量控制。OBJECTIVE To establish an HPLC method for determining the contents of palmatinechloride and berberine hydrochloride in Coptis chinensis Franch. METHODS The condition of detection were: Agilent C 18( 250mm ×4. 6mm,5μm),the mobile phase consisted of acetonitrile and 15mmol·L-1sodiumdodecylsulfate phosphate buffered saline( 0. 05mol·L-1Potassium Phosphate Monobasic,adjust with phosphoric Acid to a pH of 4. 0)( 40∶60),the FLD excitation wavelength was 354nm,the emission wavelength was 533nm. RESULTS The calibration curves were linear in the range of 1. 798 ~ 179. 8μg·mL-1for palmatinechloride( r = 1. 000); The calibration curves were linear in the range of 1. 762 ~ 176. 2μg·mL-1for berberine hydrochloride( r = 0. 9995). The average recoveries of palmatinechloride were 99. 98%( RSD = 3. 8%); the average recoveries of berberine hydrochloride were 99. 12%( RSD = 1. 4%). CONCLUSION The method is simple,accurate,reliable and can be used for the quality control of palmatinechloride and berberine hydrochloride in Coptis chinensis Franch.
