摘 要:目的:对辽宁、吉林和河南等地HIV/AIDS患者记忆性调节性CD4+CD25+Foxp3+CD45RO+T细胞水平及与疾病进展相关性进行研究,探讨记忆性调节性T细胞在HIV感染疾病进程中发挥的作用。方法:选取74名HIV感染者(缓慢进展组、无症状HIV感染组、艾滋病组、抗病毒治疗组)及15例健康对照,应用流式细胞仪检测CD4+CD25+Foxp3+CD45RO+记忆性调节性T细胞表达水平,分析其与CD4+T细胞数量、病毒载量、淋巴细胞活化及凋亡水平的相关性。结果:HIV感染缓慢进展组及抗病毒治疗组CD4+CD25+Foxp3+CD45RO+调节性T细胞水平[(2.19±1.07)%,(2.66±1.78)%]明显低于无症状HIV感染组(4.05±2.30)%,艾滋病组(5.85±2.46)%,P<0.05,无症状HIV感染组(4.05±2.30)%及健康对照(3.18±1.17)%明显低于艾滋病组(5.85±2.46)%,P<0.05。未治疗HIV/AIDS患者CD4+CD25+Foxp3+CD45RO+记忆性调节性T细胞百分率与CD4+T细胞显著负相关(r=-0.690,P<0.001),与病毒载量明显正相关(r=0.401,P=0.002),与CD4+HLA-DR+、CD8+HLA-DR+、CD8+CD38+细胞显著正相关(P<0.05),与CD4+CD95+、CD8+CD95+T细胞水平显著正相关(P<0.05)。结论:HIV感染者CD4+CD25+Foxp3+CD45RO+记忆性调节性T细胞百分率明显升高,与疾病进展及免疫损伤显著相关,是影响艾滋病疾病进展的重要因素之一。Objective:To quantify the alternations of CD4^+ CD25^+ Foxp^3+ CD45RO^ memory regulatory T ceils of HIV infec- ted patients from Liaoning, Henan and Jifin Provinces and to investigate the role of memory regulatory T cells in disease progression of HIV infection. Methods: 74 HIV infected patients and 15 normal controls were enrolled and the frequency ofCD4^+ CD25^+ Foxp^3+ CD45RO^ regulatory T cells was detected by flow-cytometry. To study the correlation between CD4^+ CD25^+ Foxp^3+ CD45RO^ regula- tory T cells and disease progression, the absolute CIM ~ T ceils, viral load and activation, apoptosis of T cells. Results: We found that the frequencies of CIM ~ CD25 + Foxp3 + CIMSRO + regulatory T ceils in slow progressors (2.19 ± 1.07 )% and patients received anti retrovi,'al therapy( 2.66 ± 1.78 )% were significantly lower than that in asymptomatic HIV infected patients (4.05± 2.30)% and AIDS patients (5.85 ± 2.46)%, P 〈 0.05. The frequency ofCD4^+ CD25^+ Foxp^3+ CD45RO^ regulatory T cells was inversely related to CD4^+ T ceils and related to viral loads, HLA-DR, CD95 expression on CIM, CD8 + T cells (P 〈 0.05). Conclusion: The frequency of CD4^+ CD25^+ Foxp^3+ CD45RO^ memory regulatory T cells was significantly increased in HIV infection and closely related to disease progression and immune responses, which indicate that alternation of memory regulatory T cells may play important role in HIV disease progresston.
