摘 要:目的观察氯胺酮对兔离体气管平滑肌细胞内游离钙离子浓度([Ca2+]i)的影响及其机制。方法采用酶解方法获得兔的气管平滑肌细胞。用钙敏荧光探针FLuo-3/AM标记细胞。将标记好的细胞随机分为氯胺酮组(A组)、氯胺酮+2-氨乙基硼酸二苯酯(2-APB)组(B组)和氯胺酮+斯里兰卡肉桂碱(RyD)组(C组)。每组再均分为三份,加入氯胺酮浓度分别为0、100和1 000μM。激光共聚焦显微镜下记录各组的[Ca2+]i。结果与氯胺酮为0μM时比较,氯胺酮为1 000μM时A、C组[Ca2+]i明显降低(P<0.05)。与A组比较,氯胺酮为0、100μM时B、C组[Ca2+]i均明显降低,氯胺酮为1 000μM时C组[Ca2+]i也明显降低(P<0.05)。结论 100μM氯胺酮对气管平滑肌[Ca2+]i无影响,1 000μM氯胺酮可以明显降低气管平滑肌[Ca2+]i。氯胺酮降低[Ca2+]i可能与其抑制内质网1,4,5-三磷酸肌醇(IP3)通路有关,与内质网兰诺定通路无关。Objective To observe the effect of ketamine on rabbit tracheal smooth muscle cells' intracellular calcium ([Ca^2+ ] i) and its possible mechanisms. Methods The single rabbit tracheal smooth muscle cells were got by enzymolysis methods. The cells were signified with FLuo-3/AM; and then were randomly divided into ketamine group (group A), Ketamine+2-APB group (group B) and ketamine+ryanodine group (group C) ; According to the density of ketamine, each group was fractionated into three subsets, that was 0, 100 and 1000 M group respectivel. The concentration of [Ca^2+]i in each group was recorded under microscope. Results In group A and C, the concentration of[Ca^2+ ]i significantly decreased when ketamine was 1 000μM compared with that when ketamine was 0μM(P〈0. 05). Compared with group A, [Ca^2+ ]i in groups B and C was significantly lower when the ketamine concentration was 0 or 100 μM, and was lower in group C when ketamine was 1 000μM (P〈0. 05). Conclusion 1 000μM ketamine can decrease the intracellular calcium, while 100 μM cannot. The above effect of ketamine may be due to block the IP3 path way, with no relationship with ryanodine way.
