摘 要:从健康的撑×绿杂交竹上分离、筛选得到抗杂交竹梢枯病菌(Arthrinium phaeospermum)的强拮抗菌株YB15。根据形态特征、生理生化特性及16S rDNA序列分析,鉴定该菌株为贝莱斯芽孢杆菌(Bacillus velezensis)。通过硫酸铵分级沉淀、Sephadex G-50分子筛层析、High Q Sepharose Fast Flow强阴离子交换层析和Sephadex G-75分子筛层析,从发酵液中分离纯化出一种分子质量约为31.0 ku的抗菌蛋白。稳定性试验显示该抗菌蛋白对蛋白酶K和胰蛋白酶不敏感,耐热,不耐酸碱和紫外线,对有机溶剂敏感性存在差异。An antagonistic bacterial strain YB-15 against Arthrinium phaeospermum was isolated from Bambusa pervaria- bilis x Dendrocalamopsis grandis. The strain was identified as Bacillus velezensis by the morphological, physiological and biochemical characteristics, and 16SrDNA sequence analysis. One of the antibacterial protein was purified by using am- monium sulfate precipitation, the Sephadex G-50 chromatography, High Q Sepharose Fast Flow chromatography and Sephadex G-75 chromatography. The molecular weight of the antibacterial protein was estimated to be 31.0 ku by SDS-PAGE analysis. Stability test showed that the antibacterial protein was resistant to proteinase K and trypsin, thermal stable, sensitive to pH and ultraviolet, had different sensitivity to organic solvents.
