摘  要:利用采自山东章丘、济宁表现典型粗缩病症状的玉米植株样品进行ELISA和RT-PCR检测。间接ELISA结果表明,二者均与水稻黑条矮缩病毒(RBSDV)多克隆抗体表现阳性反应。RT-PCR扩增出该病毒基因组S10约1.8 kb片段。获得的1 801 bp全长序列与已发表的相关序列进行同源性分析和系统进化树分析,结果表明,章丘分离物克隆JNZ-14序列、济宁分离物克隆JN-8序列均属于RBSDV,但其与玉米粗缩病毒(MRDV)的进化关系比其与南方水稻黑条矮缩病毒(SRBSDV)的关系还要近,且2个分离物分别处于属于RBSDV的Ⅱ或Ⅰ两大分支中。Typical rough dwarf symptom from Zhangqiu and Jining of Shandong was detected by ELISA and RT-PCR methods. The results from indirect ELISA indicated that, both the samples showed positive reaction with RBSDV polyclonal antibody. RT-PCR amplified about 1.8 kb segment of the viral genome S10. the 1.8 kb full-length sequence acquired had made homology and phylogenetic tree analysis with published sequences related. The results showed that, the Zhangqiu isolate clones JNZ-14 sequence, the Jining isolate clones, JN-8, belongs to Rice black-streaked dwarf virus(RBSDV),but the evolutionary relationship with MRDV were even closer than theirs with southern rice black-streaked dwarf virus(SRBSDV), moreover, the two isolates were in largeⅡorⅠbranches of RBS-DV respectively.
