摘 要:目的研究莪术挥发油中莪术二酮、吉马酮在大鼠体内的药动学特征。方法采用水蒸汽蒸馏法提取莪术中挥发油灌胃大鼠并采集血样。选用EliteHypersilODS2色谱柱(4.6mm×250mm,5μm),乙腈一水为流动相进行梯度洗脱,体积流量1.0mL/min,检测波长214nm。结果血浆样品中莪术二酮、吉马酮无干扰,分别在1.073—68.64μg/mL、0.1972—12.62μg/mL范围内呈现良好的线性关系,精密度、稳定性、回收率均符合生物样品测定要求。结论所建测定方法操作,适用于莪术二酮、吉马酮在大鼠体内血药浓度的测定及药动学两者均符合二室模型。AIM To study pharmacokinetic characteristics of curdione and germacrone in zedoary turmeric oil (ZTO) in rats. METHODS ZTO was extracted by vapour distillation, rat plasma samples were collected after oral administration. The chromatographic separation was achieved with a linear gradient elution of acetonitrile in water with a flow rate at 1.0 mL/min on a Hypersil ODS -2 column (250 mm 4.6 ram, 5 m). Detection was carried out using a UV detector at 214 nm. RESULTS Curdione and germacrone in plasma samples had no inter- ference. The linear range for curdione and germacrone were 1. 073 - 68.64 g/mL and 0. 1972 - 12.62 Ixg/mL, respectively. Precision, stability and recoveries were in line with the requirements of methodology in vivo. CON- CLUSION The developed method can be applied to the determination of curdione and germacrone in rats plas- ma. Pharmacokinetic study reveals that distribution of curdione and germacrone in ZTO is fit in with two-compart- ment model.
