摘 要:目的评价蒲黄-五灵脂配伍前后有机酸、黄酮类成分的溶出变化,以探讨药对配伍的科学性与合理性。方法采用HPLC-PDA方法同时分析测定样品中没食子酸、原儿茶酸、香草酸、对香豆酸、香蒲新苷和异鼠李素-3-O-新橙皮糖苷的量,采用Diamonsil C18色谱柱,梯度洗脱的方法进行分析测定。结果蒲黄、五灵脂配伍后香蒲新苷、异鼠李素-3-O-新橙皮糖苷溶出增加,均高于蒲黄、五灵脂单煎液和单煎混合液;没食子酸、原儿茶酸、香草酸、对香豆酸4个有机酸成分经配伍后溶出量低于单煎液,但高于单煎混合液。结论本实验建立了同时测定蒲黄-五灵脂中有机酸和黄酮类成分的HPLC-PDA方法,为揭示其效应物质基础提供科学依据与参考。AIM To evaluate the influence of compatibility of Puhuang ( Typhae Pollen) and Wulingzhi ( Tro- gopterori Faeces) on the changes in dissolution of active substances. METHODS Four organic acids and two fla- vonoids were analyzed simultaneously by HPLC-PDA. A Diamonsil Cls column ( 250 mm x 4.6 ram, 5 pm ) was used with methanol and 0. 5% acetic acid solution as mobile phase in gradient elution manner. RESULTS This method showed a good linearity for six components in their respective range and the average recoveries fell between 95.54% and 105.35% , with RSD within 1.78% - 3.07%. As a result, the contents of typhaneoside and isoham- netin-3-O-neohesperidoside increased after compatibility while the contents of gallic acid, protocatechuic acid, va- nillic acid, and p-coumaric acid, decreased. CONCLUSION The method for simultaneous determination of or- ganic acids and flavonoids in compatibility of Typhae Pollen and Trogopterori Faeces can be used in claritying the bioactive components research.
