摘 要:目的:探讨重组腺病毒介导血红素加氧酶-1基因(Adv-HO-1)对大鼠小肠缺血再灌注损伤的影响。方法:建立大鼠缺血再灌注模型,分为Adv-HO-1和生理盐水组。采用RT-PCR检测HO-1 mRNA含量,ELASA检测血清中肿瘤坏死因子α(TNF-α)、二胺氧化酶(DAO)、D-乳酸水平,免疫组织化学、Western-Blot和RT-PCR检测Occludin的含量。结果:与对照组相比,实验组HO-1 mRNA含量明显升高,TNF-α、DAO、D-乳酸明显降低,小肠病理损伤明显减轻,Occludin蛋白含量和mRNA水平明显升高,具有统计学意义。结论:HO-1能降低肠黏膜屏障的通透性并修复紧密连接、保护肠道。Objective To study the effects of recombinant adenovirus-mediated heme oxygenase-1 gene transfection on intestinal ischemia-reperfusion injury in rats. Methods The intestinal ischemia-reperfusion injury models were constructed in rats, which were divided into two groups: Adv-HO-1 and normal saline. The level of HO-1 mRNA was detected by RT-PCR. Diamine oxidase, D-lactate and TNF-c were tested by ELISA. The tight junction protein occludin was measured by immunohistochemistry, Western blot and RT-PCR. Results Compared with the control group, the level of HO-1 mRNA significantly increased while the level of Diamine oxidase, D-lactate and TNF-a significantly decreased and the expression of occludin protein was higher in the experimental group. Conclusion HO-1 can protect the intestine from ischemia-reperfusion injury in rats by improving intestinal barrier permeability and repairing tight junctions.
