摘 要:目的研究白藜芦醇对博来霉素所致小鼠肺损伤的保护作用及其机制。方法将雄性ICR小鼠随机分为对照组、博来霉素组、白藜芦醇组、博来霉素+白藜芦醇组,每组10只。小鼠气管内滴注博来霉素(3mg/kg)制作肺损伤模型,白藜芦醇(0.3mg/kg)在滴注博来霉素前注射于小鼠腹腔内。24h后取肺组织行H-E染色进行组织病理学观察,用试剂盒检测氧化应激指标丙二醛(MDA)含量,炎症因子髓过氧化物酶(MPO)活性,炎症因子IL-6、IL-8、IL-10水平。结果白藜芦醇治疗可以改善小鼠肺组织因博来霉素所致的损伤性病理变化。经白藜芦醇治疗后,博来霉素诱导的氧化应激反应得到抑制,脂质过氧化生物标记物MDA含量减少(P<0.01);博来霉素导致的肺部炎症反应减轻,MPO活性降低(P<0.01),IL-6、IL-8水平下降(IL-6:P<0.01;IL-8:P<0.05),IL-10水平增加(P<0.01)。结论白藜芦醇对博来霉素所致的小鼠肺损伤有一定的保护作用,作用机制可能与其抑制氧化应激及炎症反应有关。Objective To study the protective effects of the resveratrol on bleomycin-induced lung injury in mice and its underlying mechanism. Methods Male ICR mice were randomly divided into four groups (n=10) : control group, bleomycin group, resveratrol group, and bleomycin+resveratrol group. Mice were anesthetized and infused intratracheally with 3 mg/kg bleomycin to create lung injury model. Resveratrol (0.3 mg/kg) was administered intraperitoneally before bleomyein infusion. The severity of pulmonary injuries was evaluated 24 h after inducing lung injury. H-E staining was used to assess the lung pathology. The levels of lipid peroxidation marker malondialdehyde (MDA), neutrophil infiltration marker myeloperoxidase (MPO) activity, and cytokines IL-6, IL-8, and IL-10 were determined by commercial kits. Results Resveratrol treatment improved the bleomyein-induced lung injury in mice by significantly inhibiting the oxidative stress and decreasing lipid peroxidation marker MDA level (P^0.01). Resveratrol treatment also attenuated the bleomycin-induced pulmonary infection by significantly decreasing MPO activity (P〈0.01) and the production of pro-inflammatory eytokines IL-6 (P〈0.01) and IL-8 (P〈 0.05) and increasing anti-inflammatory cytokine ILl0 production (P〈0.01). Conclusion Resveratrol can protect against bleomycin- induced lung injury in mice, probably through its antioxidant and anti-inflammatory effects.
