摘 要:目的探讨盐酸纳美芬对大鼠颅脑损伤后脑水肿的影响。方法将54只Wistar大鼠按随机数字表法随机分为假手术组、颅脑损伤组及纳美芬治疗组。采用自由落体法建立大鼠颅脑损伤模型,分别于伤后6 h、1 d、3 d、7 d,采用干湿重法检测脑组织含水量,应用免疫组织化学法检测损伤脑组织中水通道蛋白4(AQP4)的表达。结果与假手术组比较,颅脑损伤组和纳美芬治疗组的脑组织含水量及AQP4水平明显升高(P<0.05);与颅脑损伤组相比,纳美芬治疗组脑组织含水量及AQP4表达水平明显降低(P<0.05)。通过相关性分析发现,大鼠颅脑损伤后脑组织AQP4表达水平与脑含水量呈明显正相关性(r=0.676,P<0.01)。结论盐酸纳美芬可能通过抑制AQP4表达,减轻脑水肿,发挥神经保护作用。ObjectiveTo explore the effect of nalmefene on cerebral edema of rats after traumatic brain injury(TBI).Methods Fifty-four adult Wistar rats were randomly divided into three groups,i.e.,sham-operation group(n=6),TBI group(n=24)and nalmefene-treated group(n=24).The TBI model was established using Feeney's method.The cerebral tissues were obtained one day after injury in sham-operation group.The injured cerebral tissues were obtained 6 hours,1 day,3 days and 7 days after injury in TBI group and nalmefene-treated group with 6 rats in each group and each time point.The brain water content was measured by dry-wet weight method.The expressions of aquaporin-4(AQP4)in the injured cerebral tissues were evaluated using the optical density obtained from the immunochemical staining.ResultsBoth the brain water content and the optical density of AQP4 were significantly higher in TBI group at every time point than that in sham-operation group(P〈0.05),but they were significantly lower in nalmefene-treated group at every time point than those in TBI group(P〈0.05).The expression of AQP4 was positively significantly correlated with the brain water content(r=0.676;P〈0.01).ConclusionsThe AQP4 may be involved in the development of cerebral edema in adult rats after TBI.Nalmefene can reduce the cerebral edema by down-regulation AQP4 in adult rats after TBI.
