摘 要:目的体外观察不同浓度镁离子下成纤维细胞和成骨细胞的生长情况.筛选出适宜这两种细胞生长的镁离子环境,为医用镁合金在体内降解速度的控制提供依据。方法取兔肌腱细胞和兔扁骨细胞原代培养,经传代和鉴定后,在不同浓度镁离子培养液中分别培养,观察和比较在镁离子浓度分别为0、20、50、70、100、110、120、130、140、150mmol/L时成纤维细胞和镁离子浓度分别为0、30、40、50、60、70、80、90、100、110、120mmol/L时成骨细胞的生长情况。结果成纤维细胞在镁离子浓度为0~110mmol/L时生长良好,镁离子浓度为70mmol/L时细胞活力最高,与其他浓度比较差异均有统计学意义(P〈0.05);成骨细胞在镁离子浓度为0~70mmol/L时生长良好,镁离子浓度为30mmol/L时细胞活力最高,与其他浓度比较差异均有统计学意义(P〈0.05)。结论成纤维细胞和成骨细胞的镁离子安全生存浓度分别为0—110mmol/L和0—70mmol/L。两种细胞的最适镁离子生存浓度分别为70mmol/L和30mmol/L。通过调控镁离子浓度可调控成纤维细胞和成骨细胞的生长和发育,为医用镁合金在体内降解速度的控制提供参考。Objective To find out the safe and optimal concentrations of magnesium ions tbr the growth of fibroblasts and osteoblasts, with the intention of using the data to control the degradation rate of medical magnesiom implants. Methods Tendon and skull cells were isolated from adult New Zealand rabbits for primary culture. After subculture and identification, rabbit fibroblasts were prepared for culture in the medium containing magnesium ions at concentrations of 0, 20, 50, 70, 100, 110, 120, 130, 140 and 150 mmol/L respectively while rabbit osteoblasts in the medium containing magnesium ions at concentrations of 0, 30, 40, 50, 60, 70, 80, 90, 100, 110 and 120 mmol/L respectively. Growth of the 2 kinds of ceils in different concentrations of magnesium ions was observed and compared. Results The fibroblasts grew well in a range of concentrations from 0 mmol/L to 110 mmol/L. The cell viability was the highest at the concentration of 70 mmol/L and significantly higher than those at other concentrations ( P 〈 0.05) . The osteoblasts grew well in a range of concentrations from 0 mmol/L to 70 mmol/L. The cell viability was the highest at the concentration of 30 mmol/L and significantly higher than those at other concentrations ( P 〈 0.05). Conclusions For rabbit fibroblasts the safe range of concentrations of magnesium ions is from 0 mmol/L to 110 mmol/L and the optimal concentration 70 mmol/L. For rabbit osteoblasts the safe range of concentrations of magnesium ions is from 0 mmol/L to 70 mmol/L and the optimal concentration 30 mmol/L. These data provide evidence for better controlling the in vivo degradation rate of medical magnesium implants, for we can regulate the growth of osteoblasts and fibroblasts through regulating the concentrations of magnesium ions.
