摘  要:目的:研究神经递质多巴胺对肿瘤单核细胞性髓源性抑制细胞(MDSC)抑制功能的干预作用及其对肿瘤生长的影响。方法:体内注射多巴胺,观察对肺癌小鼠模型肿瘤生长的影响;采用流式细胞术检测多巴胺注射对荷瘤鼠骨髓单核细胞性MDSC数量及细胞表型的影响;以T细胞增殖实验检测多巴胺处理对MDSC抑制功能的影响。结果:多巴胺注射可延缓肿瘤生长、明显降低单核细胞性MDSC对T细胞增殖的抑制作用,并促进MDSC的分化成熟。结论:神经递质多巴胺可干预肿瘤单核细胞性MDSC的抑制功能,可能是其抗肿瘤作用的机制之一。Objective: To investigate the interference effect of neurotransmitter dopamine on the suppressive function of tumor monocytic myeloid-derived suppressor cell ( MDSC ) and to explore its possible mechanism . Methods:By the use of lung cancer mouse model , we observed the effect of dopamine administration on the tumor growth .Proliferation assay was performed to assess whether dopamine treatment could modulate suppressive function of tumor-derived monocytic MDSC on T cell proliferation .The influence of dopamine on the quantity and phenotype of monocytic MDSC from tumor bone marrow was determined by flow cytometry .Results:Dopamine administration retarded tumor growth and reduced suppressive effect of monocytic MDSC on T cell proliferation , and also promoted its differentiation and maturation .Conclusion: Neurotransmitter dopamine can modulate the suppressive effect of tumor-derived monocytic MDSC , which may be one possible mechanism contributes to its anti-tumor activity .
