摘 要:目的研究头顶一颗珠(TTM)注射液对D-半乳糖所致小鼠衰老模型的影响。方法昆明种小鼠(雌雄各半)40只,随机分为对照组、模型组、7.2g/kg TTM组、3.6g/kg TTM组及1.8g/kg TTM组,各8只。腹腔注射D-半乳糖造模,Morris水迷宫测量小鼠学习和记忆能力。检测各组小鼠皮肤羟脯氨酸、心脑组织MDA、LP、SOD及GSH-Px含量,采用双抗体夹心ELISA法检测小鼠血清中IL-2及IL-6含量。结果 Morris水迷宫实验中,TTM各组潜伏期与模型组相比较明显减少(P<0.05),模型组小鼠穿越平台的次数比其它各组小鼠少(P<0.05)。心、脑组织匀浆氧化性指标(SOD、MAD、LP及GSH-Px)的检测,对照组及TTM各组MAD与LP含量低于模型组(P<0.05)。对照组及TTM各组SOD与GSH-Px的活性明显高于模型组(P<0.05)。注射过D-半乳糖小鼠皮肤羟脯氨酸含量都明显低于对照组(P<0.05),TTM组小鼠皮肤中羟脯氨酸的含量明显高于模型组(P<0.05)。TTM组小鼠血清中IL-2、IL-6含量显著高于对照组和模型组(P<0.05),模型组小鼠血清中的IL-2、IL-6比对照组要低(P<0.05)。结论 TTM的有效成分具有多种生物学活性,可以通过不同途径发挥抗衰老作用,在改善学习记忆功能、清除体内自由基抗氧化、增强机体免疫能力等方面都有较好的作用。Objective To study the effect of trillium tschonoskii maxim(TTM)injection on mice-aging model induced by D-galactose.Methods 40 Kunming mice(20 male and 20 female) were randomly divided into control group,model group,7.2g/kg TTM group,3.6g/kg TTM group and 1.8g/kg TTM group with eight mice in each group.The model induced by D-galactose injected peritoneal cavity and Morris water maze was used to detect the learning and cognitive ability of mice.The levels of MDA,LP,SOD and GSH-Px of skin hydroxyproline,heart and brain tissue were detected,the IL-2 and IL-6 levels in serum of mice were detected by double antibody sandwich ELISA method.Results 3 TTM groups were significantly reduced in latency period compared with the model group during Morris water maze test(P0.05) and the number of mice in model group through the platform was less than other mice in each group(P0.05).The levels of MAD and LP of the control group and each TTM group were less than model group in the detection of heart,brain tissue oxidation index(SOD,MAD,LP and GSH-Px),P0.05.The activity of SOD and GSH-Px in the control group and each TTM group was significantly higher than that in the model group(P0.05).The skin hydroxyproline content of mice injected D-galactose was significantly lower than that of the control group(P0.05) and the skin hydroxyproline content of mice of TTM group was significantly higher than that the of model group(P0.05).The IL-2,IL-6 levels in serum of mice in TTM group were significantly higher than that in the control group and the model group(P0.05) and the IL-2,IL-6 levels in serum of mice in the model group was lower than that of the control group(P0.05).Conclusion The effective constituents of TTM have a variety of biological activity which have a good effect on antiaging by different ways,improving learning and memory function,eliminating free radicals antioxidant,enhancing the body immunity and other aspects.
