养血消风饮对慢性ACD小鼠淋巴细胞凋亡及其相关细胞因子影响的实验研究

摘　　要：目的:进行养血消风饮的药效学研究,从淋巴细胞凋亡及其相关细胞因子的角度进行其免疫学机制探讨。方法:用二硝基氯苯建立小鼠耳廓慢性ACD模型,随机分为4组:空白组、模型组、中药组、西药对照组,采用免疫组化的方法,检测慢性ACD模型小鼠病变组织中细胞因子TNF-α、NF-κBp50、Fas/FasL含量,同时取小鼠脾组织冲洗液,用Annexin V-FITC细胞凋亡检测方法,用流式细胞仪检测不同组间淋巴细胞早期凋亡的百分率,并进行其与Fas/FasL含量的相关分析。通过对小鼠慢性ACD发病过程中皮损部位上述细胞因子表达的变化、脾淋巴细胞早期凋亡百分率及其相关性进行研究,探求养血消风饮可能的作用机制和环节。从细胞水平探索养血消风饮能否通过诱导淋巴细胞凋亡下调免疫应答强度,进而抑制ACD的发生。结果:养血消风饮可明显下调慢性ACD小鼠病变皮肤组织中TNF-α、NF-κBp50的表达水平,并明显上调Fas和FasL的表达,且二者分别呈正相关。同时,ACD小鼠的脾淋巴细胞早期凋亡的百分率较模型组明显增加,且与小鼠耳组织Fas、FasL的表达分别呈正相关。中药组脾淋巴细胞凋亡百分率优于西药对照组。结论:养血消风饮可明显下调慢性ACD小鼠病变皮肤组织中TNF-α、NF-κBp50的表达水平,可能在转录水平抑制炎症的发生,抑制免疫反应;同时,可明显上调其病变皮肤组织中Fas和FasL的表达,促进小鼠脾淋巴细胞凋亡。Objective ： To test the efficiency of the herbal medicine Yangxue Xiaofeng Drink. Methods ： Chronic ACD mice model stimulated on its auricles was built up by DNCB, and then all the mice were separated into four groups ： blank model group, mode[ group, traditional Chinese medicine treatment group, and western medicine control group. The con- tents of TNF -α, NF - KBp50, Fas / Fas - L of chronic lesions of cytokines in ACD mice model were detected by using immunohistochemical methods. The levels of percentage of early apoptosis of lymphocytes in mice spleen were measured. Then the correlation of TNF -/NF - KBp50, Fas / Fas - L and relationship between Fas/Fas - L and percentage of early apoptosis of lymphocytes were analyzed. Results ： Yangxue Xiaofeng Drink can significantly lower TNF -α, NF - KBp50 expression in chronic skin lesions of mice, and markedly increase that of Fas and FasL. TNF -α/NF - KBp50 and Fas/ Fas - L expression in ear skin of mice were positively correlated. Yangxue Xiaofeng Drink may significantly increase per- centage of early apoptosis of lymphocytes in the mouse spleen. And the percentage of early apoptosis of lymphocytes was positively correlated with Fas/Fas - L expressions in ear skin of mice. Collusion ： Yangxue Xiaofeng Drink can signifi- cantly lower the TNF - α, NF - KBp50 expression levels and promote Fas / FasL expression level. And it can promote apoptosis of lymphocytes in mice spleen.
