摘  要:目的:观察快速性心律失常大鼠针刺血清对乳鼠心肌细胞内Ca2+的影响,以探讨针刺治疗心律失常的离子通道机制。方法:取新生SD大鼠心肌细胞进行原代培养,5 d后应用荧光分子探针Fluo-3AM染色,应用激光共聚焦技术分别观察正常大鼠血清、模型大鼠血清和针刺大鼠血清对乳鼠心肌细胞Ca2+浓度变化的影响。结果:与空白对照组相比,模型血清组和针刺血清组Ca2+平均荧光强度值均增加,差异有统计学意义(P<0.01);与模型血清组相比,针刺血清组Ca2+平均荧光强度值降低,差异有统计学意义(P<0.05)。结论:快速性心律失常大鼠血清中有某种活性物质可以增加心肌细胞内Ca2+浓度;而针刺降低了血清中该物质含量,从而起到治疗快速性心律失常的作用。Objective : Observe the effect of acupuncture serum on the myocardial intracellular Ca2 + in tachy arrhythmia rats, explore the mechanism of acupuncture treatment of arrhythmia ion channel. Methods : Take newborn SD rat myocar- dial cells to culture the original generation, after five days with the fluorescent molecular probes Fluo -3AM staining, confocal laser technology application respectively to observe the effect of normal rat serum, model rat serum and acupunc- ture rat serum of Ca2+ concentration changes in rat myocardial cells. Results:Compared with blank control group, model serum and acupuncture serum group average fluorescence intensity of Ca2+ increased, the difference was statistically sig- nificant (P 〈 0.01 ) ; Compared with the model serum group, acupuncture serum group average fluorescence intensity of Ca2+ reduced, the difference was statistically significant (P 〈 0. 05). Conclusion:Tachy arrhythmia in rat serum having certain active substances can increase myocardial intracellular Ca2+ concentration; acupuncture treatment decrease the matter content in serum, make the therapeutic effect of tachy arrhythmia.
