摘 要:目的:建立参麦注射液在心肌缺血患者体内的群体药动学模型,为临床个体化给药提供参考。方法:用非线性混合效应模型(NONMEM)建立参麦注射液的临床群体药动学模型,考察人口统计学指标、生理生化指标等相关因素对群体药动学参数的影响,并进行模型验证。结果:以二房室模型为基础药动学模型进行拟合,群体药动学参数θCL、θVc、θQ、θVp分别为(0.011±0.0003)L·h-1,(0.1±0.007)L,(0.016±0.006)L·h-1,(0.362±0.159)L。结论:用NONMEM软件拟合建立了的参麦注射液的群体药动学模型,经验证稳定可靠,可以用于参麦注射液的临床药代动力学研究。Objective :To construct the population pharmacokinetic model of Shenmai Injection (SMI) in patients with myocardial ischemia for individualized drug therapy in clinical practice. Method:NONMEM was employed to establish the population PK model of SMI. The effects of demography and biochemical covariates were investigated. Internal and exter- nal validations were conducted for the model. Result : A two - compartment model was confirmed as structure model of SMI, the population typical values of θcf., θvo, θQ, θvp were (0.011 ±0. 0003 ) L· h^-1, ( 0. 1 ±0. 007 ) L, (0. 016 ±0. 006)L · h^-1, (0.362 ±0. 159 )L. Conclusion:The final PPK model of SMI in patients with myocardial ischemia was established by NONMEM. The PPK model was steady and reliable.
