摘  要:目的:观察不同剂量冠心合剂对大鼠急性心肌梗死血流动力学、心梗面积边缘区细胞凋亡及蛋白表达的影响。方法:取雄性SD大鼠80只,随机分为模型组、冠心合剂高剂量组、中剂量组、低剂量组和对照组。手术后4 h测量血流动力学变化后,留取心脏标本,检测梗死面积、细胞凋亡指数以及Bax、Bcl-2、Caspase-3蛋白表达量。结果:与模型组比较,冠心合剂组可明显改善心梗大鼠血流动力学,减少心梗面积及心肌细胞凋亡指数,升高Bcl-2表达,减少Bax和Caspase3蛋白表达。结论:冠心合剂改善心梗大鼠血流动力学,抑制梗死边缘区心肌细胞的凋亡、减少心肌梗死面积,其机制与调节Bcl-2和Bax表达,减少Caspase-3蛋白的表达水平有关。Objective:To investigate the effects of Guanxin Heji on hemodynamics, infarct area and apoptosis in rats of acute myocardial infarction. Methods:80 male SD rats were randomly divided into 5 groups:model group, sham group, high -dose group, medium -dose group,low -dose group. After being administered corresponding medicine for 7 days, all groups except sham group underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models and sham group only received thoracotomy without coronary ligation. Four hours after AMI, hemody- namic pararmeters were detected and myocardium tissues were collected to detect the infarct area, apoptotic index with TUNEL and protein expression of Bcl - 2, Bax and Caspase - 3 by western blot. Results : The hemodynamic pararmeters were improved in Guanxin Heji group than model group. Infarct area and apoptotic index were reduced in Guanxin Heji group than that in model group, and protein expression of Bcl - 2 was increased and Bax and Caspase - 3 were decreased significantly in Guanxin Heji group than those in model group. Conclusion:Guanxin Heji improved hemodynamic pararme- ters, reduced infarct area, inhibited the myocardial apoptosis by regulating Bel -2, Bax and Caspase -3 expression in rats of acute myocardial infarction.
