摘 要:目的:观察糖络宁对STZ诱导糖尿病大鼠糖、脂毒性及神经电生理的影响。方法:采用一次性腹腔内注射大剂量链脲佐菌素(STZ,60 mg·kg-1)诱导糖尿病大鼠模型。将糖尿病模型鼠随机分为模型组、糖络宁组、α-硫辛酸组(每组23只),正常鼠(15只)为正常对照组。糖络宁组予糖络宁按生药20 g·kg-1·d-1ig,α-硫辛酸组予α-硫辛酸20 mg·kg-1·d-1ig,其他2组大鼠予等体积蒸馏水ig,连续处理12周。观察体质量(BM)、空腹血糖(FPG)、糖化血红蛋白(HbA1c)、游离脂肪酸(FFA)水平以及摆尾温度阈值(TTT)、坐骨神经运动神经传导速度(MNCV)、感觉神经传导速度(SNCV)的变化。结果:与正常鼠比较,各组糖尿病大鼠FPG、HbA1c、FFA水平及TTT均明显升高(P<0.01),而BM、MNCV、SNCV明显下降(P<0.01);经糖络宁、α-硫辛酸处理后两组大鼠FPG、HbA1c、FFA及TTT、MNCV、SNCV均明显改善(P<0.05),但BM无明显变化。糖络宁组大鼠在FPG、HbA1c、FFA改善方面与α-硫辛酸组比较有统计学差异(P<0.05),其余指标均未见明显统计学差异(P>0.05)。结论:中药糖络宁能有效改善糖尿病大鼠的糖、脂毒性和神经传导速度,保护神经组织,对糖尿病周围神经病变(DPN)有防治作用。Objective: To observe the effects of Tangluoning (TLN) on glucose - toxicity, lipotoxicity and neuro - elec- trophysiology of streptozocin (STZ) induced diabetic rats. Methods : Diabetes mellitus was induced by a single intraperito- neal injection of large -dose streptozotocin (STZ,60 mg· kg^- 1 ) in male SD rats. Sixty -nine diabetic rats were random- ly assigned to the model group,Tangluoning (TLN) group and alpha lipoic acid (ALA) group (23 cases in each group). 15 normal rats were randomly set as normal control group. Rats in TLN group were performed intragastric administration of 20 g · kg^- 1 · d^ - 1 crude drug of TLN and rats in ALA group were administrated with 20 mg · kg^- 1 · d^ - 1 ALA, and oth- er two groups were administrated with distilled water by equal volume. All four groups had been treated for twelve weeks. Body mass ( BM), fast plasma glucose ( FPG), glycolated hemoglobin ( HbA1 c), free fatty acid ( FFA), tailflick threshold temperature (TTT) ,motor nerve conduction velocity (MNCV) and sensory nerve conduction veloc (SNCV) of rats were measured after twelve weeks. Results:Compared with normal control group, the levels of FPG, HbA1 c, FFA and TTT of all diabetic rats were significantly increased (P 〈 0.01 ) and the levels of BM, MNCV, SNCV were significantly reduced ( P 〈 0.01 ). Compared with the model group, the levels of FPG, HbA1 c, FFA,TTr, MNCV, SNCV of rats in both TLN group and ALA group were all significantly improved (P 〈 0.05 ) but BM had no significant difference (P 〉 0.05 ). The effect of TLN group on FPG, HbA1 c and FFA were superior to those of ALA group, the differences were all significant (P 〈 0. 05) , but effects on other indices had no significant difference ( P 〉 0.05 ). Conclusion : TLN can effectively improve glucose toxicity,lipotoxicity and nerve conduction velocity of diabetic rats.
