摘  要:采用毛细管电泳法测定了兔血清中胡芦巴碱含量,并对胡芦巴碱在兔子体内的药物代谢动力学过程进行了研究。试验选择了以下分析条件:①运行液为pH5.6的柠檬酸盐缓冲溶液;②检测波长为224nm;③分离电压为22kV;④进样时间为10S;⑤毛细管温度为25。C。胡芦巴碱的质量浓度在0.1~100mg·L^-1范围内与相应的峰高呈线性关系,检出限(3S/N)为0.05mg·L^-1。方法用于兔血清样品分析,采用PKS程序对血药浓度与给药时间之间的曲线进行拟合,得出系列药代动力学参数。结果表明:胡芦巴碱在家兔血液中的代谢过程符合二室开放模型特征,是一种中速吸收,快速消除的药物。CE was applied to the determination of trigonelline in rabbit serum, and pharmacokinetics of trigonelline within rabbit body was studied. Analytical conditions were studied and following optimized parameters were given: (1) electrophoretic medium: citrate buffer solution of pH 5.6; (2) wavelength for detection: 224 nm; (3) separation voltage: 22 kV; (4) time of sample introduction: 10 s';(5) temperature of capillary:25 ℃. Linear relationship between values of peak height and mass concentration of trigonelline was kept in the range of 0. 1- 100 mg · L^-1 , with detection limit (3S/N) of 0. 05 mg · L ^-1. The proposed method was used in the analysis of sample of rabbit serum, by using the PKS program to fit the plot of drug concentration in blood vs. time of drug given to obtain a series of pharmacokinetic parameters. It was found that the metabolic process of trigonelline in rabbit blood was conformed with the characteristic of two-compartment open model, and that the drug was absorbed at a middle rate and eliminated at a rapid rate.
