康复期强制戒毒人员淋巴细胞基因组DNA甲基化水平的研究

摘  要:目的:检测康复期强制戒毒人员淋巴细胞基因组DNA甲基化的水平。方法:抽提康复期强制戒毒人员和正常人淋巴细胞基因组DNA,采用整体甲基化定量试剂盒检测基因组DNA甲基化水平。结果:分别对80例康复期强制戒毒人员和正常健康人群进行淋巴细胞基因组DNA甲基化水平检测发现,与正常组相比,康复期强制戒毒人员淋巴细胞基因组DNA甲基化百分率为15.37%±s 3.28%,显著低于正常组甲基化百分率23.18%±s 7.98%,具有统计学意义(P<0.05)。不同年龄、性别组淋巴细胞基因组DNA甲基化百分率差异不显著(P>0.05)。不同吸毒年限组中,吸毒年限为>10 a组淋巴细胞基因组DNA甲基化百分率为10.01%,显著低于其他各组,差异具有统计学意义(P=0.028)。结论:康复期强制戒毒人员淋巴细胞基因组DNA甲基化水平显著低于正常人水平,吸毒时间越长,淋巴细胞基因组DNA甲基化水平降低更为显著。Objective.:To detect methylation level of genomic DNA in lymphocytes of drug addicts under compulsory rehabilitation. Methods: Genomic DNA was extracted from lymphocytes of drug users and healthy people (control). And DNA methylation levels were detected by Global DNA Methylation Quantification Ultra Kit. Results:Eighty drug users and 80 healthy people were involved in our study. DNA methylation level in lymphocytes of drug users was 15.37% ± s 3.28%, which was significantly lower than that of control(23.18%±s 7.98% ,P 〈0. 05). However, there was no significant difference among different age and sexual groups ( P 〉 0. 05 ). The DNA methylation level of drug users with drug - use history of more than 10 years was 10.01% ,which was significantly lower than that of those with less drug - use history( P = 0. 028 ). Conclusion : The methylation level of genomic DNA in lymphocyte of drug users under compulsory rehabilitation was significantly lower than that of healthy people and decreased with years of drug use.
