摘 要:【目的】研究生肌调节因子肌细胞生成素(MyoG)和生肌决定因子(Myf 6)基因表达对鸭胚胎期及初生早期骨骼肌发育的影响。【方法】采用Real-time PCR方法研究MyoG、Myf6基因在不同鸭品种(高邮鸭和金定鸭)胚胎期(13,17,21,25,27胚龄)及初生早期(出雏后7日龄)骨骼肌发育中的表达差异,并分析其表达变化与胸、腿肌发育的相关性。【结果】MyoG和Myf6基因在2个品种鸭胸肌、腿肌组织发育早期均有表达,但不同组织存在不同的表达模式。MyoG在鸭胚胸肌、腿肌组织中均呈现"前高后低"的表达模式,21胚龄以前表达量相对较高,随后表达量下降,并维持相对较低水平;而Myf6基因在胸肌组织中在21胚龄时表达量最高,随后表达量下降,但仍维持相对较高的表达水平;在腿肌组织中在13胚龄时表达量较高,在17胚龄时达到最高,随后表达量有所降低,并在21胚龄后急速下降至极低水平,但其表达在胚胎期后期(27胚龄)以及初生期(7日龄)又有缓慢上升趋势。二元变量相关性分析结果表明,2个品种鸭胸肌、腿肌MyoG mRNA表达与胸肌、腿肌发育呈强负相关;胸肌中Myf6mRNA的表达与胸肌发育无显著相关性,腿肌中Myf6mRNA的表达与腿肌发育呈强负相关。【结论】骨骼肌中MyoG和Myf6基因表达具有显著的时空性,性别差异不显著,骨骼肌中MyoG和Myf6基因表达与鸭发育早期骨骼肌质量变化呈现负的线性相关。【Objective】Effects of MyoG and Myf6 genes on the skeletal muscle in early development stage of duck were studied.【Method】The mRNA expressions of MyoG and Myf6 genes on the skeletal muscle of Gaoyou duck and Jinding duck (Anas platyrhynchos domestica) during embryonic stage on days 13,17,21,25,27 and 1 week after patch were detected using quantitative RT-PCR.【Result】MyoG and Myf6 genes were detected in early development stage of both Gaoyou duck and Jinding duck with different expression patterns.The expression of MyoG gene on both pectorals and legs showed a relatively high expression level on days 13,17,and 21,and then the expression level declined to a low level on days 25,27 and 1 week after patch.The expression of Myf6 gene on pectorals reached the highest level at the embryo age of 21 day,and subsequently the expression decreased to a slightly lower level.Expression level of Myf6 gene on legs at embryo ages of 13 and 17 was high,and then it decreased slightly until the embryo age of 21,when it declined rapidly to the lowest level.The expression of Myf6 gene on legs in the late embryonic stage (day 27) and the newborn period (within 1 week after birth) increased slightly.Bivariate correlation analysis showed that the expression of MyoG gene and pectorals weight and legs weight were negatively correlated;while the expression of Myf6 gene was negatively correlated with legs weight.【Conclution】MyoG and Myf6 genes involved in the skeletal development,and different expression patterns in pectorals and legs suggested that MyoG and Myf6 genes may be related to chest and leg muscle development and differentiation.
