摘 要:目的调查柳州地区献血人群中HBV的基因型分布和不同基因型HBV的群体动力学。方法采集来自广西血液中心2009年HBV确证阳性的献血者血浆标本,用半巢式PCR的方法扩增HBV基因组S区的序列,测序后通过贝叶斯推断探讨柳州地区分布的HBV基因型;应用系统发育学和群体遗传学方法,对不同的HBV基因型群体进行群体遗传学分析,探讨该地区不同基因型HBV的扩张情况。结果柳州地区献血者中HBV有3种基因型,分别是B型(71.4%)、C型(26.5%)和D型(2%)。B型和C型HBV群体的Tajima's D检验与Fu's Fs检验结果都为负值,同时这2个群体的错配分布也呈现单一峰倒钟形分布,说明柳州地区的HBV群体都在扩张。贝叶斯轮廓图推断B型和C型HBV目前有相似的有效种群大小,但起源时间B型晚于C型。结论柳州地区献血者中HBV基因型多样性较丰富,B型和C型的HBV都正在经历1个缓慢扩张的过程,需持续研究和监控HBV在献血者中的感染情况和遗传分化情况,做好相应的血液预警工作。Objective To investigate the genetic typing and population dynamics of the Hepatitis B virus discovered in blood donors from Liuzhou Blood Center. Methods Plasma samples were collected in 2009. Semi-nested PCR was used to amplify the DNA fragment located in the S region of tlBV genome. Bayesian Inference was employed to reveal the diversity of HBV genotypes in Liuzhou, and the population structure was also investigated under population dynamics analysis. Results There were 3 genotypes of HBV distributed in blood donors from Liuzhou:genotype B (71.4%) ,genotype C (26. 5% ) and genotype D (2%). Negative values of Tajima's D and Fu's Fs,as well as unimodal mismatch distributions suggested the HBV population of genotype B and genotype C underwent recent expansions. The Bayesian Skyline Plot results also suggested the slow expansions and a relatively close population size of HBV populations of genotype B and genotype C. Meanwhile,the ori- gin time of genotype B in Liuzhou was estimated earlier than genotype C. Conclusion Abundant HBV diversity was found in Liuzhou. The HBV in this region, both genotype B and genotype C,were undergoing slow expansions. So, continuously research and surveillance on the population genetics of HBV in blood donors should be executed to enhance the blood safety in Liuzhou area.
