清肝化痰活血方对非酒精性脂肪性肝炎大鼠肠上皮细胞紧密连接蛋白occludin表达的影响

摘　　要：目的:观察清肝化痰活血方对非酒精性脂肪性肝炎(NASH)大鼠肠上皮细胞紧密连接蛋白occludin表达的影响。方法:将40只SD大鼠随机分为正常组、模型组、易善复组和清肝化痰活血方组(简称清肝组)。采用高脂饲料复制NASH模型大鼠;清肝组和易善复组在进食高脂饲料的同时分别予清肝化痰活血方药液和易善复混悬液进行灌胃;第12周时处死所有大鼠,留取血清和肠组织标本。采用全自动生化分析仪检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST);双抗体夹心ABC-ELISA法检测肿瘤坏死因子-α(TNF-α)的含量;经HE染色光镜下观察肠黏膜组织的病理学变化;RT-PCR法检测肠组织中紧密连接蛋白occludin的mRNA表达。结果:模型组大鼠血清AST、ALT、TNF-α的含量与正常组比较明显升高(P<0.01),肠上皮细胞紧密连接蛋白occludin mRNA的表达与正常组相比明显下降(P<0.01);清肝组大鼠血清AST、ALT、TNF-α的含量较模型组降低(P<0.05,P<0.01),肠组织中的occludin mRNA的表达较模型组明显增加(P<0.01)。结论:清肝化痰活血方具有改善和修复NASH大鼠肠黏膜损伤的作用。Objective： To investigate the effect of ＂ Qinggan Huatan Huoxue Decoction＂ on occludin expression of enterocyte connexin in rats with nonalcoholic steatohepatitis （NASH）. Methods： Forty SD rats were randomized into normal group, model group, Essentialc group and ＂Qinggan Huatan Huoxue Decoction＂ group （Qinggan group）. The models were established by giving high fat diet. Qinggan group and Essentiale group were treated with ＂Qinggan Huatan Huoxue Decoction＂ and Essentiale respectively by intragastric administration. Rats were sacrificed at 12 weeks after modeling, the serum and intestinal mucosa samples were kept. The levels of ALT and AST were detected by automatic biochemical analyzer; contents of TNF-α were detected by ABC-ELISA; the pathologic changes of intestinal mucosa were observed by HE staining; the expression of occludin mRNA was detected by RT-PCR. Results： The serum levels of AST, ALT, TNF-α in model group were increased and higher than that of the normal group （ P 〈 0.01 ） , and the occludin proteins levels of intestinal mucosa samples were decreased and lower than that of the normal group （P 〈 0.01 ） ; the contents of AST, ALT and TNF-α in Qinggan group were lower than that of the model group （P 〈0.05, P 〈0.01 ）, and the expression of occludin mRNA was higher than that of the model group （P 〈 0.01 ）. Conclusion： ＂ Qinggan Huatan Huoxue Decoction＂ can improve and repair the damaged intestinal mucosa in NASH rats.
