西格列汀对肥胖MSG大鼠糖代谢和胰岛局部血管紧张素Ⅱ的影响

摘　　要：目的:探讨DDP-Ⅳ抑制剂西格列汀对肥胖MSG大鼠糖代谢和胰岛局部血管紧张素Ⅱ(Ang-Ⅱ)的影响。方法:大鼠分为正常对照组(Con组)、MSG组(出生后接受左旋谷氨酸单钠注射,成年后高脂饮食)、MSG+L组(MSG鼠氯沙坦灌胃干预)、MSG+S组(MSG鼠西格列汀灌胃干预),行OGTT、胰岛素释放试验及ITT评估大鼠血糖、胰岛β细胞功能及胰岛素抵抗情况,ELISA法检测系统和胰腺局部Ang-Ⅱ水平。结果:与Con相比,MSG组大鼠体重、脂肪湿重、Lee′s指数升高,血糖升高,血浆胰岛素水平下降,且胰岛素敏感性降低;与MSG组相比,MSG+S组血糖水平下降、胰岛素水平升高、胰岛素抵抗改善,胰腺Ang-Ⅱ水平降低,而MSG+L组虽然血糖水平下降、胰岛素抵抗改善,仅基础胰岛素水平升高。结论:DDP-Ⅳ抑制剂改善MSG大鼠胰岛β细胞功能及胰岛素抵抗,其机制与胰腺局部Ang-Ⅱ表达下降有关。Objective To investigate the effects of sitagliptin on glycometabolism and pancreatic local rennin-angiotensin system （RAS） in obesity rats induced by monosodium glutamate （MSG）. Methods Rats were assigned into normal control group, MSG group （injected with MSG, fed with high-fat diet）, MSG ＋ L group （treated with losartan）, MSG ＋ S group （treated with sitagliptin）. Oral glucose tolerance test, insulin release test and insulin tolerance test were performed for evaluating the glucose status including insulin sensitivity and [~ cell functions. System Ang-II and local Ang-II in pancreatic islets were detected by ELISA. Results As compared to those in control group, the body weight, Lee＇s index, wet weight of peritoneal adipose tissue and blood glucose in MSG group were increased; but serum level of insulin and insulin sensitivity were decreased. As compared to those in MSG group, blood glucose and local Ang-II were decreased, and serum level of insulin and insulin sensitivity were increased in MSG ＋ S group; whilst blood glucose was decreased, and serum level of insulin and insulin sensitivity were increased in MSG ＋ L group. Conclusion Sitagliptin could improve insulin resistance and β cell functions in the obesity rats induced by MSG, which may be related to the down-regulation of local Ang- II in pancreatic islets.
