摘  要:目的研究脱细胞人胎羊膜(HAM)对大鼠皮肤全层切口愈合过程中转化生长因子β(TGF-β)及凝血酶敏感蛋白1(TSP-1)表达的影响,探讨其促进全层切口愈合及无瘢痕愈合的发生机制。方法建立Sprague-Dawley大鼠全层皮肤缺损模型,然后将大鼠分为对照组和HAM组,每组15只。对照组大鼠切口用生理盐水冲洗后纱布包裹,HAM组大鼠切口外敷HAM并用纱布包裹。应用免疫组织化学技术分析切创后不同时相大鼠皮肤组织中TGF-β及TSP-1的表达。结果切口处理后第3天对照组大鼠皮肤组织TGF-β及TSP-1表达高于HAM组,差异有统计学意义(P<0.05)。结论 HAM可减少大鼠皮肤全层切口早期愈合过程中TGF-β及TSP-1的表达,促进切口愈合,减少瘢痕增生。Objective To study the effect of acellular human amniotic membrane(HAM) on the expression of transfor- ming growth factor β (TGF-β) and thrombospondin-1 ( TSP-1 ) during full-thickness skin wound healing process of rats ; and ex- plore the mechanism of HAM in promoting full-thickness skin wound healing. Methods The full-thickness skin wound models of Sprague-Dawley rats were established, then the rats were divided into control group and HAM group, with fifteen rats in each group. The incisions of rats in control group were wrapped with gauze ; the incisions of rats in HAM group were applied with HAM and then wrapped with gauze. The immunohistochemistry method was used to analyse the expression of TGF-13 and TSP-1 in the incised skin tissue at different phases. Results At the third day after incised,the expressions of TGF-β and TSP-1 in control group were significantly higher than those in HAM group (P 〈 0.05). Conclusion HAM can reduce the expression of TGF-β and TSP-1 in the early full-thickness skin wound healing of rats and promote wound healing and reduce scar.
