摘 要:目的研究不同浓度氟对大鼠血清氧化应激效应的影响。方法采用饮水中加入不同剂量的氟化钠饲喂大鼠复制氟中毒大鼠模型,饲喂90 d和120 d后,各组随机处死5只大鼠,检查氟斑牙发生情况,采用比色法检测血清中总抗氧化能力(T-AOC)、黄嘌呤氧化酶法测定血清中超氧化物歧化酶(SOD)活性,硫代巴比妥酸法测定血清中丙二醛(MDA)含量。结果 90 d和120 d时,各染氟组大鼠出现不同程度的氟斑牙,且随染氟剂量的增大和染氟时间的延长而加重。低氟组两时间点大鼠血清中T-AOC[(9.37±5.82)U/ml,(11.31±2.90)U/ml]均低于相应的对照组[(22.76±7.99)U/ml,(21.37±6.43)U/ml],差异有统计学意义(P<0.01或P<0.05)。各组之间SOD活性差异均无统计学意义。90 d时,中氟组和高氟组MDA含量[(8.08±2.72)nmol/ml,(9.67±1.70)nmol/ml]均明显高于对照组[(4.26±0.53)nmol/ml],差异均有统计学意义(P<0.01);120 d时,各染氟组MDA含量[(7.49±0.47)nmol/ml,(10.48±1.97)nmol/ml,(12.50±2.59)nmol/ml]均明显高于对照组[(4.70±0.63)nmol/ml],差异均有统计学意义(P<0.05或P<0.01)。结论慢性氟中毒可导致大鼠血清氧化系统和抗氧化系统失衡,氧化应激在氟中毒中可能发挥重要作用。Objective To study the effect of fluoride at different dosages on the oxidative stress in rat serum. Methods Forty male SD rats were randomly divided into control group, low fluorosis group, middle fluorosis group and high fluorosis group, with 10 rats in each group. Rats in three fluorosis groups were orally given drinking water with various concentrations of sodium fluoride. After 90 and 120 days, five rats in each group were randomly sacrificed to observe the dental fluorosis. And the total anti - oxygen capacity ( T - AOC), the activity of superoxide dismutase (SOD) and the content of malonaldehyde (MDA) in serum were measured by biochemical methods. Results Different degrees of dental fluorosis were found in all fluorosis groups, showing positive correlation with the experimental time and fluorine doses. T - AOC level [ (9.37 ± 5.82) U/ml, ( 11.31 ± 2.90) U/roll in low fluorosis group was significantly lower than that in the control group[ (22.76 ± 7.99) U/ml, (21.37 ± 6.43 ) U/roll at two time points, the difference was statistically significant(P 〈0.01 or P 〈0.05), but there were no signifi- cant differences of SOD among different groups on the 90th and 120th day. The contents of MDA [ (8.08 ± 2.72 ) nmol/ml, (9.67 ± 1.70) nmol/ml] in the middle fluorosis group and high fluorosis group were significantly higher than those in the control group [ (4.26 ± 0.53 ) nmol/ml ] on the 90th day ( P 〈 0.01 ). The contents of MDA [ (7.49 ± 0.47 ) nmol/ml, ( 10.48 ± 1.97) nmol/ml, (12.50 ± 2.59) nmol/ml] in all fluorosis groups were significantly higher than those in the control group [ (4.70± 0.63 ) nmol/ml ] on the 120th day( P 〈 0.05 or P 〈 0.01 ). Conclusion Chronic fluoride poisoning can cause disorders of the oxidative system and anti - oxidative system in rat serum. The oxidative stress may play an important role in fluorosis.
