摘　　要：目的探讨肺结核患者的血清中血管内皮生长因子(VEGF)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达与变化。方法对疑似肺结核病患者采样痰涂片抗酸染色法和酶联免疫反应法检测筛选检测,在确诊为肺结核患者时,静脉采血,通过Real-time RT-PCR法和Western-blot检测其血清中VEGF、TNF-α、IL-6 mRNA和蛋白表达量的变化与正常健康人进行比较。结果通过对95例疑似结核患者筛选,确诊有63例肺结核患者,通过Real-time RT-PCR检测发现VEGF、TNF-α、IL-6 mRNA的表达量明显著高于正常健康人组,Western-blot检测结果表明肺结核患者体内VEGF、TNF-α、IL-6蛋白表达量显著高于对照组。结论结核患者体内VEGF、IL-6表达水平升高,通过检测研究VEGF、TNF-α、IL-6表达水平的变化可研究结核杆菌对人体致病机制。Objective To discuss the expression and changes of vascular endothelial growth factor （VEGF）, interleukin 6 （IL -6） and tumor necrosis factor-α（ TNF - ct） in tuberculosis patients. Methods Sputum samples from suspected pulmonary tuberculosis patients were detected by smear acid -fast staining and enzyme -linked immunosorbent assay, then the confirmed pulmonary tuberculosis patients were conducted venous blood collection to detect the serum VEGF, TNF -α, IL - 6 mRNA and protein expression by real - time RT - PCR and Western blot, and the detection results were compared with those of normal healthy individuals. Results In 95 suspected tuberculosis patients, 63 cases were confirmed as pulmonary tuberculosis, and the mRNA expression levels of VEGF, TNF -α and IL - 6 were significantly higher than those in normal control group. Western blot test results showed that patients with pulmonary tuberculosis had higher protein expression levels in VEGF, TNF -α, IL - 6 than those of control group. Conclusion The expression levels of VEGF and IL - 6 increase in tuberculosis patients, and the detection of VEGF, TNF -α, IL -6 expression level changes contribute to study the pathogenic mechanism of mycobacterium tuberculosis on human.
