摘 要:目的构建表达乙型肝炎病毒 (HBV) S蛋白的重组质粒 p CR3.1- S作为核酸疫苗 ,观察重组人白细胞介素 2(rh IL - 2 )作为佐剂对其诱导 BAL B/ c小鼠产生免疫应答的影响。方法分别以 EL ISA法、3H- Td R掺入法、Cr5 1 4h释放法等测定免疫小鼠的血清抗 HBs、淋巴细胞增殖活性、淋巴细胞杀伤效应 ,初步研究不同免疫组体液和细胞免疫应答的差异。结果血清抗 HBs EL ISA效价 ,rh IL - 2组与对照组相比较差异不显著 (P>0 .0 5 ) ;淋巴细胞增殖指数、淋巴细胞杀伤率 ,10 0 0 U (只·次 ) rh IL - 2 /组和 2 0 0 0 U (只·次 ) rh IL - 2 /组与对照组相比较差异显著 (P<0 .0 5 ) ,而 5 0 0 U (只·次 ) rh IL - 2 /组与对照组相比较无显著差异 (P>0 .0 5 )。结论一定剂量的 rh IL - 2作为佐剂可增强ObjectiveTo construct recombinant plasmids of pCR3 1 S encoding protein S of hepatitis B virus as nucleic acid vaccines and investigate the effects of recombinant human interleukin 2 as adjuvants on immune responses induced by HBV nucleic acid vaccines in BALB/c mice Methods Anti HBs antibodies to HBV major protein was measured by ELISA assay , and the activities of lymphocyte proliferation and cytotoxity were detected by[KG-2/3] 3H TdR incorporation assay and 4 hour 51 [KG2/3]Cr release assay Humoral and cellular immune responses of different groups were then compared Results The titers of anti HBs antibodies in serum of rhIL 2 group mice were not significantly higher than those of control group mice ( P >0 05). The diffe rence of the activities of lymphocyte proliferation and cytotoxity between of control group mice and rhIL 2 group mice (every mouse with 500 U one time) was not significant ( P >0 05). However, stimulation index and specific lysis rate of lymphocyte of rhIL 2 group mice (every mouse with 1 000 U or 2 000 U one time) showed significant difference compared with htose of control group mice ( P <0 05). Conclusion Recombinant plasmids of pCR3 1 S as HBV nucleic acid vaccines could induced strong humoral and cellular immune responses in BALB/c mice rhIL 2 acts as adjuvants to increase the potency of HBV nucleic acid vaccine in some certainty
