摘 要:目的 探讨备用根大鼠手术侧和非手术侧脊髓后角组织 >10 k D和 <10 k D两种分子量组分提取液 ,对鸡胚背根节 (DRG)神经突起的促生长作用 ,以及应用吗啡后对此作用的影响。 方法 应用超滤器制备提取液、组织培养和 SDS-聚丙烯酰胺凝胶电泳等技术 ,分离和检测神经营养活性物质。 结果 备用根大鼠手术侧脊髓后角提取液 >10 k D组分作用的 DRG神经突起的生长密度 ,比非手术侧 >10 k D组分 ,非手术侧 <10 k D组分和手术侧 <10 k D组分的生长密度明显增大。应用吗啡的备用根大鼠手术侧脊髓后角组织 >10 k D组分提取液作用的 DRG神经突起生长密度 ,又比备用根大鼠手术侧 >10 k D组分的大。经 SDS-聚丙烯酰胺凝胶电泳分析发现 ,两组动物手术侧和非手术侧 >10 k D组分样品的电泳图谱区带数目大致相同 ,但两组动物手术侧分子量约 6 7k D、31k D和 16 k D的 3条区带吸收峰面积百分比 ,较非手术侧的高。 结论 吗啡具有进一步增强去传入纤维支配的脊髓后角组织 >10 k D组分提取液促进 DRG神经突起的生长效应。两组动物手术侧 >10 k D组分提取液分子量约 6 7k D、31k D和16 k D3种蛋白质含量有增高的趋势 ,尤其是吗啡组动物手术侧约 6 7kObjective The neurite\|promoting effects of two molecular weight fractions(>10kD and <10kD) from the extracts of operated side and unoperated side spinal dorsal horn in spared root rat on dorsal root ganglion (DRG) of chick embryo were explored. The influence of the extracts from morphine spared root rat on the neurite\|promoting effects was also observed. Methods The neurotrophic active substances were isolated and analyzed by some methods of ultrafilter preparing the extract, tissue culture and SDS\|polyacrylamide gel electrophoresis (SDS\|PAGE). Results DRG neuritic density treated by molecular weight fraction of >10kD from the extract of operated side spinal dorsal horn in spared root rat was larger than that of>10kD and <10kD from the extract of unoperated side, and of <10kD from the extract of operated side. The DRG neuritic density treated by molecular weight fraction of >10kD from the extract of operated side spinal dorsal horn in morphine spared root rat was again larger than that of operated side spinal dorsal horn in spared root rat. The number of electrophoretic zone of samples was approximately the same in operated side and unoperated side, whereas the percentage of absorptive peak area of three zones with approximate 67kD, 31kD and 16kD from operated side of two groups of animal was larger respectively in comparison with unoperated side. Conclusion Morphine might enhance the neurite\|promoting effect of the extract of >10kD fraction from deafferented spinal dorsal horn on DRG. The content of 67kD, 31kD and 16kD protein at >10kD fraction from operated side of two groups of animal was tending towards increasing, especially the approximate 67kD protein content from operated side of morphine group of animal.\;
