调节肝纤维化模型大鼠TGF-1表达探讨雷帕霉素改善肝纤维化的相关性

摘  要:目的:研究雷帕霉素调节肝纤维化模型大鼠转化生长因子1(transformation growth factor 1,TGF-1)的表达,改善肝纤维变的相关作用.方法:应用大鼠以四氯化碳油(CCl4花生油)诱导建立肝纤维化模型,具体分成正常对照组、肝纤维化模型组、RAPA组,饲养8 wk后观察动物肝脏组织形态学改变及TGF-1在肝组织的表达.在动物饲养过程中动态地测定肝纤维化进程中血清TGF-1水平的变化.结果:3组大鼠肝组织切片经HE及免疫组织化等染色后,图片经Image-pro plus 6.0图像分析软件整理.A(IA值9891.4±1725.7)、B(IA值为104239.9±20890.6)、C(IA值为27853.6±2031).3组大鼠肝组织切片中TGF-1表达呈显著性差异,具有统计学意义(P<0.01).结论:雷帕霉素具有显著的下调肝脏组织中TGF-1表达及下调血清TGF-1水平,TGF-1表达下调与改善肝纤维化变有平行关系.METHODS: Rats were divided into three groups: a normal control group, a liver fibrosis model group and a rapamycin treatment group. Rats were treated with carbon tetrachloride to induce liver fibrosis. Eight weeks after treat-ment, liver histological changes and the expres- sion of TGF-β1 in the liver were detected. In addition, serum TGF-β1 levels were dynamically monitored. RESULTS: After immunohistochemical and HE staining, image-pro plus 6 image analysis soft- ware was used to analyze the integrated absor- bance (IA). The IA values were 9891.4 ± 1725.7, 104239.± 20890.6, and 27853.6 ± 2031 in the normal control group, liver fibrosis model group and rapamycin treatment group, respectively. The expression of TGF-β1 in liver tissue was sig- nificantly different among the three groups (P 〈 0.01). CONCLUSION: Rapamycin can significantly re- duce the levels of TGF-β1 in liver tissue and se- rum, which may be involved in the therapeutic effect of rapamycin on liver fibrosis.
