摘 要:目的探讨血清学检测方法和实时荧光定量PCR方法在新型布尼亚病毒检测中的应用。方法连续采集烟台地区莱州、蓬莱疑似发热伴血小板减少综合征患者血清,分别用胶体金实验方法、ELISA实验方法检测抗新型布尼亚病毒IgM抗体,实时荧光定量PCR方法检测新型布尼亚病毒核酸,SPSS18.0统计结果。结果胶体金方法和ELISA方法检测结果总符合率为96.67%,一致性KAPPA检验K系数为0.933;胶体金方法和实时荧光定量PCR方法检测结果均受病程影响(两者P值均小于0.05);实时荧光定量PCR2疗法检测不受年龄、性别影响(P均〉0.05)。结论新型布尼亚病毒检测发病12天以前推荐使用实时荧光定量PCR方法,发病16天以后,可使用血清学方法检测抗新型布尼亚病毒IgM抗体。Objective To compare serological methods with real time RT-PCR in detection of severe fever with thrombocytopenia syndrome Bunyavirus. Method Serum of severe fever with thrombocytopenia syndrome was collected in Penglai and Laizhou in Yantai. Anti-severe fever with thrombocytopenia syndrome Bunyavirus (SFTSV) IgM was detected using gold colloid and ELISA, nucleic acid of severe fever with thrombocytopenia syndrome Bunyavirus was detected by real-time PCR. Statistics of the results were done using SPSS 18.0. Result The coincidence of ELISA and gold colloid was 96.97% and the kcoefficient was 0. 933 ; the course of disease influences the results notablely both gold colloid and real-time PCR(P 〈 0. 05 ) and real-time PCR was not influenced by patients age or sex ( P 〉 0. 05 ). Conclusion When the course of disease less than 12 days, real-time PCR was recommended and when the course of disease more than 16 days, serological methods can use to detect the anti-SFTSV IgM.
