摘  要:分析简单节杆菌乙醇耐性差异菌株的生长特性,以静息细胞为对象,考察乙醇处理对菌体细胞存活率的影响,进一步分析细胞糖代谢能力、细胞膜电位、细胞超显微结构和通透性的变化.结果发现:乙醇体积分数在0%。10%的范围内,突变菌株UV15X1.2比出发菌株TCCC11037表现出更好的生长特性;6%和10%乙醇处理后,突变菌株UV15X1—2的细胞存活率比出发菌株TCCC11037分别高出19.2%和18.3%,表现出更好的乙醇耐受性;进一步研究发现,6%乙醇引起出发菌株TCCC11037糖代谢能力和膜电位的明显下降,细胞完整性遭到明显破坏,而对于突变菌株UV15X1.2,这些变化在乙醇体积分数为10%的条件下出现.此外,通过测定FDA的相对荧光强度和细胞蛋白质及核酸的泄漏量,表明一定体积分数的乙醇处理能显著提高简单节杆菌的细胞通透性.The growing properties of Arthrobacter Simplex strains with different organic solvent tolerance were studied. Using resting cells as materials, the effects of ethanol on the cell survival, sugar metabolic activity, membrane potential, cell ultrastructure and permeability were investigated. The results showed that the growing properties of UV15X1-2 were better than those of TCCC 11037 within the range of ethanol concentration (0%-10%) . After the treatment with 6% and 10% of ethanol, the cell survival rate of UV15X1-2 was 19.2% and 18.3% higher compared with TCCC 11037, which suggested that UV15XI-2 presented higher tolerance toward ethanol. As for TCCC 11037, sugar metabolic activities and membrane potential decreased rapidly, and the cell integrity was significantly damaged with 6% of ethanol, but similar effects on cell properties and structure were not observed until UV15X1-2 was treated with 10% of ethanol. On the other hand, the relative fluorescence intensity of FDA and the protein/nucleic acid leakage revealed that ethanol could enhance the permeability of the Arthrobacter simplex cell significantly.
