摘 要:目的研究康艾冻干粉针剂体内外抗肿瘤细胞的活性。方法建立小鼠S180、H22肿瘤模型,测定不同浓度康艾粉针溶液体内对荷瘤小鼠肿瘤的抑制作用。同时进行体外实验,选取小鼠肿瘤细胞S180、人肝癌细胞HepG2,分别以与不同浓度的康艾粉针溶液体外共同培养作为给药组,与5-Fu共同培养作为阳性对照组,并设空白对照组。MTT法检测待测药物对肿瘤细胞的抑制作用,荧光显微镜下观察药物对HepG2细胞核形态学特征的影响,采用流式细胞仪对HepG2细胞的细胞周期时相及凋亡率进行分析。结果康艾粉针在给药剂量为3~12g/kg时,对S180、H22荷瘤小鼠肿瘤均产生明显的体内抑制作用,且在该剂量范围内呈现一定的量效关系,对小鼠S180肿瘤的抑制率为20.20%~54.55%,对H22肿瘤的抑制率为30.18%~68.65%。康艾粉针对小鼠S180、人肝癌细胞HepG2的增殖均有明显的抑制作用,且具有直接杀死HepG2以及诱导HepG2凋亡的双重作用。结论康艾冻干粉针剂具有良好的体内外抗癌活性。Objective To examine the in vivo and in vitro anti-tumor activity of Kang'ai freeze-dried powder.Methods The tumor-bearing models were established in S180 and H22mice.The Kang'ai freeze-dried powder at different concentrations was injected into the tumor-bearing mice to observe the anti-tumor effect.Meanwhile,the in vitro experiments were performed.The S180 and HepG2 cell lines were cultured and divided into the drug group(in which the cells were treated with different concentrations of Kang'ai freeze-dried powder),positive group(in which the cells were treated with 5-Fu)and blank control group(in which no drugs were given).MTT assay was used to measure the proliferation of the cells.The morphological features of the nuclei of the HepG2 cells were observed under the fluorescent microscope and FCM was employed to detect the HepG2cell apoptosis rate and the change of cell circle.Results Kang'ai freeze-dried powder injected at 3-12g/kg significantly inhibited the growth of tumors in a dose-dependant manner in S180and H22mice.The anti-tumor rate was 20.20%-54.55% in S180 mice and 30.18%-68.65%in H22 mice.Moreover,the Kang'ai freeze-dried powder had significant inhibitory effects on the proliferation of the S180 and HepG2cells.It could both directly kill the HepG2cells and induce the apoptosis of HepG2 cells.Conclusion The Kang'ai freeze-dried powder has good anti-tumor activities in vivo and in vitro.
