摘 要:目的探讨染色体着丝粒点(Cd)的变异与JAR细胞染色体非整倍性畸变的关系。方法肿瘤细胞株传代后用常规方法制备染色体,用直接法制备胚胎绒毛组织染色体标本,采用Cd-核仁组织区(NOR)同步银染分析技术研究JAR细胞染色体Cd的变异。结果 JAR细胞染色体Cd缺失率为1.33%、Cd迟滞复制率为1.23%、小Cd率为1.73%、Cd-NOR融合率为0.62%,与健康人胚胎绒毛细胞染色体Cd相比较,JAR细胞染色体Cd缺失、Cd迟滞复制显著升高(P<0.0125),而小Cd、Cd-NOR融合其差异无统计学意义。结论 JAR细胞染色体非整倍性畸变可能主要源于Cd缺失、Cd迟滞复制。Objective To study the relation between the centromeric dots(Gd)variation of JAR ceils and the chromosome aneuploidy aberration. Methods Chromosome of tumor cells were prepared with general method,and chromosome of normal embryonic villi were prepared straightly. A Cd-NOR in-phase argentum dye analysis tech- nique modified by this laboratory was used to study centromeric dots(Cd) variation of JAR cells. Results Fre- quency of Cd loss in JAR cells was 1.33%. Frequency of Cd duplication laggard was 1.23~. Frequency of little Cd was 1.73%. Frequency of Cd-NOR amalgamation was 0.62%. Frequency of Cd loss and Cd duplication lag- gard in JAR cells were significantly higher than those of normal embryonic villi cells (P^O. 0125). Frequency of little Cd and Cd-NOR amalgamation showed no differenee between JAR cells and normal embryonic villi. Conclu- sion Cd loss and Cd duplication laggard might be related with aneuploidy aberration of JAR cells.
