摘 要:目的:观察不同时间点聚乳酸(OPLA)/基质胶(matrigel matrix,MM)在裸鼠体内的促血管生成情况,评价其促血管生成的效果。方法:以OPLA复合支架为载体,分为处理组和对照组。处理组:OPLA+MM;对照组:OPLA。选取3只雄性裸鼠,2种支架分别植入每只裸鼠的前肢与背部交接处,1、2、4周取材,进行HE染色和免疫组织化学染色,观察OPLA/MM促血管生成的效果,计算微血管密度(MVD),采用独立样本t检验法进行定量分析,比较其MVD的差异。结果:1~4周内随着时间的延长,支架与周围组织融合良好,移植物周围的血管逐渐增多,并有肉眼可见的血管长入;每个时间点OPLA+MM的MVD值较高;同时,OPLA支架随时间延长逐渐降解,支架内部被新生组织分割成小区域,并伴随有血管长入。结论:在裸鼠体内,OPLA+MM具有良好的促血管生成作用。Objective To explore the angiogenesis effects of OPLA/Matrigel Matrix (MM) at different timepoints and to analyze the microvwssel density (MVD). Mt^laocls Open-cell polylatice acid (OPLA) covered with MM and OPLA (control) was implanted into null-mice. These scaffolds were harvested at the timepoints of 1, 2 and 4 weeks, and then HE and immunocytochemistry was employed to detect the angiogenesis promoting effect of scaffold, Results The results showed that the scaffold had good biocompatibility with the tissue. A great number of vessels were observed obviously surrounding the implants, especially at the timepoint of week 4. The OPLA scaffolds degraded with time going by. MVD of OPLA+MM was higher than OPLA at any timepiont. Coneluaion Combined OPLA with MM shows a good angiogenesis prompting effect in null-mice.
