摘 要:目的:应用PCR-STR技术,研究干细胞移植患者外周血、口腔黏膜上皮细胞及毛根的基因型变化。方法:对44例干细胞移植患者的血液、口腔拭子及毛根进行分型。结果:44例患者移植后外周血检出单一供者基因分型,毛根均检出单一受者基因分型,口腔黏膜上皮细胞有16例检出单一受者的基因分型,28例检出供者、受者的混合基因分型。结论:移植后外周血干细胞可以转变为口腔黏膜上皮细胞,这对干细胞治疗各种血液恶性疾病及法医学应用具有较高的参考价值。Objective: To investigate the genotype diversity of the periph-eral blood cells,epithelial cells from oral mucosa and root-hair cells in re-cipients following transplantation of hematopoietic stem cells from the pe-ripheral blood.Methods: Polymerase chain reaction of short tandem re-peat sequences ( PCR-STR ) assay was performed for genotype of the blood,samples by oral swab and root-hair cells in 44 recipients.Results:The genotype of donor origin was commonly identified in 44 patients in samples from both peripheral blood and root-hair cells.In addition,16 of 44 subjects were appearance of recipient′s genotype for the mucosal epi-thelial cells and 28,coexistence of donor and host hematopoiesis ( mixed chimerism) .Conclusion: The transplanted allogeneic peripheral blood stem cells may convert into buccal epithelial cells , and this finding is of high value in diagnosis of engraftment failure in patients with malignancy after allogenetic stem cell transplantation and forensic case-work.
