摘 要:目的利用小鼠拘束应激负荷模型评价四逆散汤剂的疏肝解郁作用,并通过检测应激性激素水平以及和肝脏炎症、氧化水平的变化探讨其可能的作用机制。方法建立小鼠拘束应激肝郁模型,测定血浆丙氨酸氨基转移酶(ALT)、天门冬酸氨基转移酶(AST)、皮质酮(CORT)水平,检测肝组织激素受体(GR)基因表达水平、抗氧化能力指数(ORAC)、丙二醛(MDA)水平、一氧化氮(NO)水平、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果与模型组相比,四逆散能显著降低应激小鼠血浆ALT和AST水平,并能降低拘束负荷小鼠血浆CORT水平和肝组织NO及MDA水平,提高ORAC水平、SOD及GSH-Px活性。结论四逆散的疏肝解郁作用可能与缓解应激负荷引起的肝损伤有关,其保护机制与抑制应激负荷引起的炎症发生以及氧化损伤作用有关。Objective To study the effects for soothing the liver and relieving depression of Sini Powder decoction by using restraint stress model in mice, and further to study the possible mechanism of Sini Powder by testing the stress- induced hormone levels and the changes of liver inflammation and oxidation level. Methods The restraint stress liver depression model in mice was established. Plasma alanine aminotransferase(ALT), aspartateaminotransferase (AST) and corticosterone(CORT) levels were detected. The HPLC method was adopted to test the corticosterone(GC) level in plasma. Nitric oxide (NO) was detected by the Greiss method and the oxygen radical absorbance eapaeity(ORAC) in liver was determined by the automated multifunction microplate reader. MDA, SOD and GSH-PX levels in liver were also determined. The reverse transcription polymerase chain reaction (RT-PCR) method was used to detect the mRNA expression of hormone receptor(GR) in the mice liver. Results Compared with the model group, Sini Powder could significantly reduce the plasma ALT and AST levels, decreased plasma CORT level and he NO and MGA levels in liver tissue of restrained mice. Sini Powder could increase the ORAC level and SOD and GSH- Px activity. Conclusion The Sini Powder's effects for soothing the liver and relieving depression could be related with relieving the liver injury caused by restraint stress and its protective mechanism could be related with the stress- induced inflammation occurrence and oxidative injury action.
