摘 要:目的探讨粒细胞集落刺激因子(G-CSF)预处理动员干细胞系对大鼠心肌缺血再灌注损伤的早期影响及其可能机制。方法 SD大鼠30只随机分3组:①假手术组;②对照组;③动员组。动员组造模前注射G-CSF 1周后行血清学检测;3组造模后24h分别检测心肌酶谱指标(CK、CKMB、CTnI),同时应用ELISA法观察血管内皮生长因子(VEGF)在术后24h的表达;采集各组组织标本分别行组织学检测并运用RT-PCR法检测各组CD34、Sca-1、Bcl-2及CTnI的表达。最后针对动员组选取G-CSF注射前、造模前及造模后24h三个时间点检测CD34与Sca-1的表达。结果经G-CSF动员后动员组白细胞较其他两组升高[(14.80±1.21)×109/L、(7.45±0.80)×109/L、(6.96±0.89)×109/L,P<0.05);造模后24h,动员组心肌酶谱指标较对照组下降(P<0.05);动员组VEGF表达较其他两组升高[(66.95±8.28)pg/mL、(39.45±9.68)pg/mL、(47.86±5.45)pg/mL,P<0.05]。RT-PCR检测半定量分析结果显示,动员组CD34、Bcl-2较其他两组表达增加(P<0.05);动员组Sca-1较假手术组表达增高(P<0.05);动员组与假手术组CTnI表达没有统计学差异(P=0.37),但两者较对照组CTnI表达降低(P<0.05)。动员组在3个时间点检测结果显示,造模前及造模后24h的CD34及Sca-1表达较注射前高(P<0.05)。结论 G-CSF预处理可以动员干细胞系,其对大鼠心肌缺血再灌注损伤的早期有一定的保护作用,从而改善大鼠的心脏功能。Objective To explore early effects and possible mechanism of stem cell-lines mobilization with granulocyte colony stimulating factor (G-CSF) on myocardial ischemia-reperfusion injury in rats. Methods A total of 30 Sprague-Dawley rats were randomly divided into sham group, control group and mobilization group. Serological test was performed 7 d after G-CSF was injected in mobilization group. The myocardial enzyme indexes (CK, CKMB and CTnl) were detected and the serum level of VEGF was observed by ELISA in the three groups 24 h after mobilization. Tissue samples in all were collected and CD34, Sca-1, CTnI, and Bcl-2 expressions were detected 24 h after operation by RT-PCR assay. Finally, CD34 and Sca-1 expressions in mobilization group were analyzed at three time points (before G-CSF injection, 24 h before and after mobilization). Results Mobilization with G-CSF increased the level of WBC in mobilization group [(14. 80± 1.21) ×10^9/L vs. (7.45±0.80)×10^9/L and (6.96 ±0.89)×109/L, P〈0.05]. The myocardial enzyme indexes were lower in mobilization group than in control group (P〈0.05) 24 h after operation, and the serum level of VEGF was higher in mobilization group than in the other groups [(66.95~8.28)pg/mL vs. (39.45_+9.68)pg/mL and (47.86~ 5.45)pg/mL, P〈0.05]. RT- PCR assay showed that higher expressions of CD34 and Bcl-2 in mobilization group than in the other groups (P〈 0.05). Sca-1 had a higher expressed in mobilization group than in sham group (P〈0.05). CTnI expression did not differ significantly between mobilization group and sham group (P = 0.37). However, CTnI had a higher expression than in the other groups (P〈0.05). In mobilization group, CD34 and Sca-1 had higher expressions 24 h before and after mobilization than before G-CSF injection (P〈0.05).
